摘要 表面等离子共振生物传感是一种光学检测技术,可获得生物分子相互作用的特异性、亲和力和动力学常数等,无需对研究的分子进行标记、灵敏度高且可实时检测,已在生命科学的发展中发挥了重要作用,正在成为蛋白质组学研究和药物发现与开发的有力工具。本文详细回顾了该技术从物理现象发现到理论体系的完善,以及从实验室研究到仪器商品化的发展历程,概述了各个时期的研究特点、仪器化以及应用情况,探讨了有待解决的问题,并展望了未来的发展趋势。 Surface plasmon resonance biosensing is an optical technique which is capable of acquiring specificity, affinity and kinetics constants of biomolecular interaction. Owing to its high sensitivity and real time detection without labeling, it plays a more and more important role in life science and has become a powerful tool in proteomics research and drug discovery. This paper reviews the development from the discovery of the SPR phenomenon to the perfection of the theoretical system and the evolvement from laboratorial prototypes to commercial SPR instruments. The characteristic features of research, instrumentation and application in each period are summarized. Problems to be solved for SPR biosensing technique and the orosoect of future trends are also discussed.
