摘要 目的应用携带hVEGF基因的重组腺病毒载体转染大鼠骨髓基质干细胞(BMSCs),观察hVEGF的表达情况。方法体外培养大鼠骨髓基质干细胞,含hVEGF基因的重组腺病毒转染骨髓基质干细胞,转染后在荧光显微镜下观察并采用ELISA法检测hVEGF的表达水平。结果 rAd-hVEGF转染骨髓基质干细胞后,ELISA检测发现转染组上清液中hVEGF蛋白分泌量明显高于对照组(P<0.01)。结论 rAd-hVEGF转染大鼠骨髓基质干细胞能够表达并分泌hVEGF,为缺血性脑血管疾病的基因治疗提供可行性研究。 Objective To transfect rAd-hVEGF into the bone marrow stromal cells (BMSCs) of SD rats, and then to assay the expression of VEGF. Methods rAd-hVEGF transfected BMSCs were cultured in vitro. The expression of VEGF in BMSCs after transfection was determined by observing the expression of GFP. ELISA method was applied to assay the secretion of hVEGF. Results EL1SA analysis showed the quantity of expression of hVEGF in transfection group was higher than that in control group (P〈0.01). Conclusion Exogenous hVEGF can be expressed in BMSCs, which provided the basis of ischemic cerebrovaseular disease's gene therapy.
