摘要 以燕子掌叶片为外植体,在培养基中添加不同浓度的6-BA和IBA,研究燕子掌组培苗形成过程中6-BA和IBA浓度对各阶段的影响。结果表明:MS+IBA 5mg/L+6-BA 5mg/L为愈伤组织诱导的最佳培养基,分化不明显,且诱导率为95.56%;MS+IBA 2mg/L+6-BA1mg/L为外植体直接诱导不定芽的最佳培养基,平均可达12芽以上;MS+IBA 5mg/L+6-BA5mg/L为愈伤组织增殖最佳培养基,增殖倍数为10.9;愈伤组织分化的最佳培养基为MS+IBA1mg/L+6-BA 5mg/L,单位面积芽数为32个以上,且芽明显;壮苗的最佳培养基为MS+IBA2mg/L,45d后平均苗长为4.74cm;诱根的最佳培养基为MS+IBA 0.5mg/L,诱根率为100%。 Taking Crassula argentea Thunb lamina as the explant and adding different concentrations of 6-BA and IBA in the culture medium,the influence of concentration of 6-BA and IBA on each stage in the Crassula argentea Thunb tissue culture forming process were studied. The results showed that MSq-IBA 5 mg/L-+-6-BA 5 mg/L was the best medium for callus induction with uneonspieuous differentiation and an inductivity of 95. 56% ;MS-k-IBA 2 mg/L+6-BA 1mg/L was the best for adventitious bud induction directly With an induetivity of 86. 67% ,that could achieve more than 12 buds in average;MS+ IBA 5 mg/L+6-BA 5 mg/L was the best for callus proliferation and the proliferation multiples was 10. 9;tbe best for callus differentiation was MS-t-IBA 1 mg/L+6-BA 5 mg/L,the number of buds per unit area was more than 32 and the buds were obvious;when the medium was MS+IBA 2 mg/+6-BA 5 rng/L,tbe globular embryos produced more on the tissue surface with a long duration;the best for sound seeding was MS+IBA 2mg/L,the average length of buds could be 4. 74 cm after 45 d;on the medium with MS+IBA 0. 5 mg/L,tbe inductivity of root inducing could be 100%.
