摘要
谷氨酸氧化酶经固定化与过氧化氢电极构成酶电极,建立了酶电极法测定鸡精中谷氨酸钠含量的方法。结果表明,该方法对鸡精谷氨酸钠的检测快速而准确,检测时间为20 s,相对标准偏差为0.74%,加标回收率为99.3%~101.2%;在1 mg/100 mL~100 mg/100 mL范围内,酶电极传感器(SBA-40E)测定值与谷氨酸质量浓度线性关系良好,相关系数R=0.999 86;检测灵敏度高,鸡精中谷氨酸检出下限为1.27 mg/100 mL;与高效液相色谱方法、高氯酸滴定法相比较,差异不显著(P〉0.05);专一性强,不受鸡精其它添加剂的干扰;重现性和稳定性好。
Glutamate oxidase was immobilized to prepare enzyme electrode constituted with hydrogen peroxide electrode.The enzyme electrode biosensor was used to detect sodium glutamate content in chicken essence.The results showed the biosensor could detect sodium glutamate rapidly and accurately,with response time of 20 s,recovery ratio of 99.3 %-101.2 %,and repeatability relative standard deviation(RSD) of 0.74 %.The biosensor(SBA-40E) showed good linear relationship within the scope of 1 mg/100 mL-100 mg/100 mL with a correlation coefficient(R) of 0.999 86.And the detector also showed some good merits such as high precision(the limit of content of sodium glutamate in chicken essence was 1.27 mg/100 mL),good repeatability,high specificity,high speed and stability.Compared with HPLC and perchloric acid titration,determination results had not significant difference(P0.05).In addition,this method was was more suitable to detect sodium glutamate in chicken essence production.
