HBV对肝内TGF-β1蛋白表达和人肝星状细胞系LX-2活性的影响 被引量:2

摘要 目的探讨HBV对肝内TGF-β1蛋白表达及人肝星状细胞系LX-2细胞活性的影响。方法运用免疫组织化学方法检测对照组(HBV阴性)和慢性乙型肝炎组肝组织内TGF-β1的表达;采用细胞培养、Western blot、MTT和Transwell实验体外观察HBV对LX-2细胞形态及活性的影响。结果慢性乙型肝炎组肝组织内TGF-β1的表达水平显著高于对照组(t=4.2268、P=0.000);在慢性乙型肝炎组,TGF-β1在肝纤维化处于S1~S2期的肝组织内表达较高,与肝纤维化S3~S4期肝组织内的表达比较差异具有显著性(t=6.9107、P=0.000)。HBV可引起LX-2细胞的形态学改变,并增加其增殖和运动迁移能力。结论 HBV主要在慢性乙型肝炎肝纤维化形成的早期阶段发挥作用。HBV感染可引起肝组织内TGF-β1蛋白表达增加和人肝星状细胞系LX-2的形态改变和活性增强。 Objective To evaluate the effects of HBV on the expression of intrahepatic TGF-β1 and the activity of human hepatic stellate cell line LX-2 cells. Methods The expression of intrahepatic TGF-β1 in control group and CHB group were detected by immunohistochemical method. The influences of HBV on morphology and activity of IL-2 ceils were observed by cell culture, Western blot, MTr and Transwell tests. Results There was significantly stronger expression of intrahepatic TGF-β1 in patients with CHB than that in control group. In CHB group, the level of TGF-β1 protein in liver tissues with fibrosis St-Sz stages was significantly higher than that with fibrosis S3-S4 stages. HBV could change LX-2 ceils' morphological and increase the abilities of proliferation and migration. Conclusions HBV plays an important role in early stage of fibrosis formation in patients with CHB. HBV infection may induce TGF-β1 expression in hepatic tissues and change hepatic stellate cell line LX-2 cells' morphological and enhance the activity.
