Antitumor Mechanisms of extracts from L. erythrorhizon Sieb. et Zucc

摘要 紫草(Lithospermum erythrorhizon Sieb.et Zucc.)作为传统中药,其化学成份复杂,具有广泛的药理作用,抗肿瘤作用是其中之一。紫草提取物能抑制多种肿瘤细胞生长增殖,导致肿瘤细胞凋亡。其作用机制包括:下调癌基因在癌细胞中的表达、抑制基因转录、抑制DNA拓扑异构酶(DNA topoisomerase)和DNA合成、抗有丝分裂和抗血管发生、诱导活性氧(reactive oxygen species,ROS)产生、调节促分裂原活化的蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路、引起Bcl-2蛋白家族表达变化进而促进细胞色素C(cytochrome C)的释放、激活胱天蛋白酶(caspase)家族启动凋亡、致肿瘤细胞周期阻滞、影响死亡受体家族及其相关蛋白活性等。 Lithospermum erythrorhizon Sieb. et Zucc. is traditional Chinese medicine. And its chemical composition complex has extensive pharmacological actions, such as anti-tumor function. The extracts from Lithospermum erythrorhizon Sieb. et Zucc can inhibit the proliferation in variety of tumor cells, resulting in tumor cells' apoptosis. The anti-tumor mechanisms include: down regulating oncogenes in cancer cells, inhibiting gene transcription, inhibiting DNA topoisomerase and DNA synthesis, resisting mitosis and anti-angiogenesis, inducing reactive oxygen species produced, regulating MAPK signal pathway, causing Bcl-2 protein family expression change and promoting the release of cytochrome C, activating caspase protease family start ap0ptosis, arresting the tumor cell cycle, influencing death receptor family and its related protein activity and so on.
