桃胶多糖对Caco-2细胞模型吸收葡萄糖的影响 被引量:10

摘要 目的:通过Caco-2体外细胞模型研究桃胶多糖影响葡萄糖吸收的机制。方法:以培养的Caco-2单层细胞为葡萄糖吸收模型,观察在桃胶多糖影响下,葡萄糖吸收量随时间的变化关系,从而推断桃胶多糖对葡萄糖跨膜转运过程的影响。结果:一定浓度桃胶多糖作用下,不同浓度葡萄糖的吸收量与对照组相比,均有显著差异(P<0.05),表明葡萄糖吸收受到桃胶多糖的抑制,且葡萄糖浓度越低,桃胶多糖抑制葡萄糖吸收的效果越显著(P<0.05);不同浓度的桃胶多糖作用下,相同浓度葡萄糖的吸收量与对照组相比,均有显著差异(P<0.05),且桃胶多糖浓度越高,抑制葡萄糖吸收的效果越好(P<0.05)。结论:桃胶多糖能够竞争性抑制葡萄糖的吸收,其是否可能通过影响或者占据肠道刷状缘侧钠葡萄糖共转运载体1(SGLT-1)上葡萄糖的吸收位点达到降血糖目的,其机制尚有待进一步研究。 This study was aimed to investigate the possible mechanism of peach gum polysaccharide affect glucose absorption in Caco-2 cell culture model. Caco-2 cell was used as in vitro drug transport model. Under the influ- ence of peach gum polysaccharide, the relationship between glucose absorption and time change was observed in order to infer the transepithelial transport of glucose in the peach gum polysaccharide solution. The results showed that under the action of a certain concentration of peach gum polysaccharide, the absorption amount of different concentrations of glucose with the control group showed a significant difference (P 〈 0.05). It showed that the glucose absorption was inhibited by peach gum polysaccharide. And the lower the glucose concentration is, the more significant the inhibition by peach gum polysaccharide is (P 〈 0.05). Compared with the control group, dif- ferent concentrations group of peach gum polysaccharide with the same concentration of glucose showed a signifi- cant difference (P 〈 0.05). And the higher the concentration of peach gum polysaccharide is, the more significant the inhibition to the glucose is (P 〈 0.05). It was concluded that peach gum polysaccharide can inhibit glucose absorption competitively. But whether it can influence or even occupy the glucose absorption sites on the intesti- nal brush border side of the sodium glucose cotransporter-1 (SGLT-1) in order to reduce the postprandial blood glucose should still be confirmed with more researches.
