柴胡疏肝散对非酒精性脂肪肝大鼠脂质代谢及肝功能的影响

Influence of chaihushugan powder on lipid metabolism and liver function in nonalcoholic fatty liver rats

目的:探讨柴胡疏肝散对非酒精性脂肪肝大鼠降脂保肝的作用。方法:取SD大鼠随机分成正常组、模型组、东宝肝泰组(0.09g蛋氨酸/kg)、柴胡疏肝散高、中、低剂量(12.6、6.3、3.15g生药/kg)。除正常组外,其余各组ig高脂高糖乳剂复制非酒精性脂肪肝大鼠模型,造模同时给予各给药组ig相应药物,正常组、模型组ig生理盐水,连续8周后,测定各组大鼠血清血脂、游离脂肪酸(FFA)、肝功能,以及肝脏总胆固醇(TC)、甘油三酯(TG)、丙二醛(MDA)、超氧化物歧化酶(SOD)、总谷胱甘肽(T-GSH),计算肝指数,光镜下观察肝脏病理学变化。结果:柴胡疏肝散可显著降低非酒精性脂肪肝大鼠血脂、肝脂、肝功能、肝MDA水平(P<0.05、0.01),对肝指数的降低、肝SOD活性和T-GSH含量的升高呈一定的剂量变化趋势但无显著差异。肝脏病理切片显示柴胡疏肝散可明显减轻肝细胞脂肪变性、水样变性及炎症细胞浸润等病理症状。结论:柴胡疏肝散对大鼠非酒精性脂肪肝具有调节脂质代谢和降酶保肝的作用,其机制可能与其减少脂质在肝脏的蓄积、降低肝细胞脂肪变、抑制炎症和脂质过氧化有关。

To investigate the effects of Chaihushugan powder on lipid-lowing and llver-protecting of nonalcoholic fatty liver disease （NAFLD） rats. Methods：The rats were randomly divided into normal control group, model group, Dongbaogantai group （0.09g methionine/kg） ,Chaihushugan powder high,medium,low doses（12.6,6.3,3.15g/kg）. In addition to the normal group, the other group was administered by i. g high fat and sugar emulsion in the morning, four hours later, each group was interfered corresponding medicine, the control group and model group were given normal saline. The rats were fed for 8weeks. At the end of the experiment, detected blood lipid,FFA,liver function, liver total cholesterol,TG, MDA, SOD, T-GSH in each group rats, calculated the liver index, observed the liver pathological changes. Results： Compared to NAFLD rats, the levels of blood lipid, liver function, liver MDA （ P 〈 0. 05,0.01 ） were obviously decreased in Chaihushugan powder groups ; to the decreasing of liver index, the increasing of SOD and T-GSH have no significant difference; liver pathological section showed the hepatocyte adipose degeneration,water samples degeneration, cell infiltration in Chaihushugan powder groups were significantly lighter than model group. Conclusion：Chaihushugan powder have the effect of adjusting lipid metabolism,protecting liver and diminishing enzymes to NAFLD rats, the mechanism is possibly related to reduce hepatocyte adipose degeneration, anti-inflammatory and inhabit lipid peroxidation.
