摘要
目的:探讨经纳米板转导HPV16 E7siRNA对宫颈癌组织增殖和凋亡的影响。方法:建立裸鼠宫颈癌移植瘤模型,纳米板将HPV16 E7siRNA转导入肿瘤组织细胞,转导后2 d用TUNEL法检测实验组和对照组的凋亡情况,免疫组化法检测实验组和对照组Ki-67的表达情况。结果:经纳米板转导HPV16 E7siRNA后实验组细胞凋亡显著增加,Ki-67表达明显降低,与对照组相比凋亡指数、增殖指数差异均具有显著性(P<0.05)。结论:纳米板转导的HPV16 E7siRNA对宫颈癌肿瘤组织增殖具有明显的抑制作用。
Objective: To evaluate the effects of nanopatch delivering HPV16 E7siRNA on proliferation and apoptosis of cervical cancer.Methods: Set up cervical cancer nude models,with nanopatch deliveried HPV16 E7siRNA into the transplanted tumor of nude mice models;apoptosis and the expression of Ki-67 were detected by TUNEL and immunohistochemistry staining at 2 days after treatment.Results: After treatment there were many TUNEL positive cells in experimental group,the expression of Ki-67 decreased significantly,the difference of apoptosis index and proliferation index between experimental group and control group was significant(P 0.05).Conclusions: Using nanopatch to deliver siRNA to cervical cancer can induce apoptosis and inhibiting of the proliferation significantly.
