摘要
目的:探讨左西孟旦(levosimendan)对急性病毒性心肌炎(acute viral myocarditis,AVM)小鼠心功能的改善作用。方法:将C57/BL6小鼠随机分为对照组、心肌炎组及心肌炎左西孟旦治疗组,给予心肌炎组及心肌炎左西孟旦治疗组小鼠腹腔注射柯萨奇病毒以形成AVM小鼠模型,心肌炎左西孟旦治疗组予每日灌服左西孟旦溶液1 mg/(kg·d)。每日观察各组小鼠行为学改变,并于7 d后行颈动脉插管,测定心脏收缩期及舒张期心室内压最大上升及下降速率(+LVdp/dt max,-LVdp/dt max)、主动脉内收缩压(SAP)及舒张压(DAP);断颈处死后称量全心重量,计算心体比;制备心肌组织病理切片,计算炎细胞浸润及心肌坏死区与每张心肌组织切片总面积之比。结果:给药7 d后,与心肌炎组小鼠比较,心肌炎左西孟旦治疗组小鼠心肌组织炎细胞浸润及心肌坏死区面积虽无明显改变(P>0.05),但行为改变出现延迟,心体比显著下降(P<0.05),+LVdp/dt max、-LVdp/dt max、SAP及DAP均显著提高(均P<0.05)。结论:左西孟旦能够改善AVM小鼠心脏功能。
Objective To evaluate the role of levosimendan in the improvement of cardiac function in mice with acute viral myocarditis (AVM). Methods C57/BL6 mice were randomly divided into a control group, AVM group, and AVM-levosimendan group. AVM group and AVM-levosimendan group were induced by intraperitoneal injections of eoxsackievirus B3., to generate the AVM mice model. AVM-levosimendan group received oral levosimendan [ 1 mg/(kg.d)] for 7 days after infection. Behavior changes were observed as the mice were administered for 7 consecutive days. The maximum rise rate and maximum decline rate of left ventricular pressure (+LV dp/dt max and -LV dp/dt max), and systolic and diastolic arterial pressure (SAP and DAP) were detected by carotid arterial cannulation. The ratios of heart/body weight, phlogocyte infiltration and necrosis/total cardiac crosssectional area were measured after the mice were killed. Results As compared with AVM group, the ratio of phlogocyte infiltration and necrosis/total cardiac cross-sectional area did not alter in AVM-levosimendan group (P 〉 0.05) after therapy with levosimendan for 7 days. But behavior changes were delayed. The ratio of heart/body weight was decreased (P 〈 0.05). +LVdp/dt max, -LVdp/dt max, SAP, and DAP were significantly increased (P 〈 0.05). Conclusions Levosimendan can improve cardiac function in mice with acute viral myocarditis.
