人参皂甙经PI3K/AKT通路诱导乳腺癌细胞凋亡 被引量:8

摘要 目的探讨人参皂甙Rg3经磷脂酰肌醇-3激酶/蛋白激酶B(P13K/AKT)通路对人乳腺癌MCF7/Adr细胞的诱导凋亡作用。方法不同浓度人参皂甙Rg3作用于人乳腺癌MCF7/Adr细胞24~72h后,MTT法检测细胞活力;荧光显微镜下观察细胞核的形态学改变;流式细胞术检测细胞凋亡率;Westernblot法检测P13K、AKT的表达。结果人参皂甙R加对人乳腺癌MCF7/Adr细胞的生长有明显的增殖抑制作用,呈浓度和时间依赖性。经荧光染色后可观察到典型的凋亡小体,细胞凋亡率明显增高,MCF7/Adr细胞P13K、AKT表达均明显减弱。结论人参皂甙Rg3能诱导体外培养的人MCF7/Adr细胞凋亡,抑制P13K、AKT活性是人参皂甙Rg3诱导MCF7/Adr细胞凋亡的重要机制之一。 Purpose To explore the inducing effect of ginsenoside Rg3 ( GS-Rg3 ) on apoptosis of human breast cancer MCFT/Adr cells by impacting PI3K/AKT pathway. Methods Human breast cancer MCF7/Adr cells were treated with diferent concentrations of GS-Rg3 for 24 ~ 72 h, and the cell viability was measured by MTT assay, cell nucleus morphology change was observed with the fluo- rescence, apoptosis was evaluated by floweytometry. Western blot were used to detect the expressions of PI3K and AKT. Results GS- Rg3 inhibited the proliferation of MCFT/Adr cell line in a dose-and time-dependent manner. The fluorescence microscope showed clearly cell apoptosis with apoptotic body, the expression levels and activities of PI3K and AKT decreased significantly. Conclusions GS-Rg3 could inhibit MCF7/Adr proliferation through inducing cell apoptosis, inhibiting the activities of PI3K and AKT is one of the important mechanisms of GS-Rg3 inducing apoptosis of MCF7/Adr cells in vitro.
