摘要
目的探讨大肠癌中Hedgehog信号通路相关基因SMO的表达及其临床意义。方法应用RT-PCR及免疫组织化学法(Envison二步法)检测43例大肠癌组织和距离癌10 cm以上的癌旁组织的SMO mR NA及蛋白的表达情况,并与20例非肠癌患者的正常大肠组织对照,分析其与大肠癌患者临床和病理各变量的相关性。结果大肠癌组织SMO mRNA和蛋白表达阳性率分别为83.7%和60.5%,与癌旁组织的58.1%和27.9%比较差异有显著性(P<0.05),且均显著高于正常组织的0%和0%(P<0.05)。大肠癌组织中SMO mRNA和蛋白表达强度均比癌旁及正常大肠组织显著增高(P<0.05)。大肠癌组织中的SMO mRNA和蛋白表达强度与患者的年龄、性别、临床分期、肿瘤部位、肿瘤浸润深度、病理分型等变量均无明显关系(P>0.05)。结论 SMO mRNA和蛋白在大肠癌组织中的表达阳性率及强度显著增高,其表达与临床和病理各变量不相关。
[Objective] To evaluate the expressions of Hedgehog signaling pathway related gene SMO in colorectal carcinoma (CRC) tissues in order to explore its significance in CRC clinicopathology. [Methods] 43 specimens of fresh CRC tissues and adjacent tissues (10 cm beyond the cancer margin) were collected and detected for their expressions of SMO mRNA and proteins with RT-PCR and immunohistochemistry (Envision method), and compared with 20 specimens of normal colorectal tissues from non-CRC patients as controls. The correlation of SMO mRNA and proteins expressions and clinicopathological variables were analyzed. [Re- suits] The positive rates of SMO mRNA and protein expressions in CRC tissues (83.7% and 60.5%) were significantly differ from those (58.1% and 27.9%) in the adjacent tissues (P〈0.05), and significantly higher than those (0% and 0%) in normal colorectal tissues (P〈0.05). The expression intensities of SMO mRNA and protein in CRC tissues were significantly higher than those adjacent tissues and normal colorectal tissues (P〈 0.05). No significant correlation was found between SMO mRNA and protein expressions and the clinical variables such as age, gender, Dukes stages, tumor locations, depth of tumor invasion and histological types of CRC (P〉0.05). [Conclusion] The positive rates and expression intensities of Hedgehog signaling pathway related gene SMO mRNA and protein expressions increased significantly in CRC tissues. The high expressions were not related to the clinieopathological indexes of severity.
