实时定量PCR检测慢性粒细胞白血病患者Shh、Smo基因的表达水平 被引量:3

摘要 目的建立Hedgehog信号通路功能性基因Shh和功能性受体蛋白Smo基因的定量检测方法,并分析其在慢性粒细胞白血病(chronic myeloid leukemia,CML)患者中的表达水平。方法利用实时定量PCR检测20例CML患者和作为正常对照(8例)的健康人外周血细胞中Shh、Smo基因的表达水平,以β2微球蛋白(β2-M)基因作为内对照。结果 CML组Shh、Smo表达水平均明显高于正常对照组(P<0.05)。结论成功建立Shh、Smo基因的定量检测方法;Shh、Smo基因在CML中高表达。 Objective To establish Hedgehog signaling pathway detection method for Shh gene and Smo gene using real-time PCR quantitatively, and analyze their expression levels in chronic myeloid leukemia (CML) patients. Methods Using Real-time PCR with SYBR Green technique , Shh gene and Smo gene expression levels in peripheral blood mononuclear ceils from 8 normal individuals (control) and 20 CML patients was detected with 32-microglobulin (132M) as reference to acquire the relative expression levels. Results Shh and Smo expression levels in CML patients were significantly higher than those in the normal control (P 〈 0.05 ). Conclusion The quantitative detection method for Shh and Smo gene expression by real- time PCR with SYBR Green I technique has been successfully established. Shh gene and Smo gene are highly expressed in CML patients.
