小半夏加茯苓汤及其组分对H22荷瘤小鼠的抑瘤作用和脾脏caspase-3基因表达的影响 被引量:3

Antitumor effect of Xiaobanxia Plus Fuling Decoction and its components and its effects on caspase-3 mRNA expression in splenic tissue of H22 tumor-bearing mice

摘要 目的:探讨小半夏加茯苓汤(简称XBPF)及其组分对H22荷瘤小鼠的抑瘤作用和脾脏组织caspase-3基因表达的影响。方法:通过给昆明小鼠接种H22肿瘤细胞,建立荷瘤小鼠模型,将60只小鼠分为空白对照组、模型组、多糖组、姜辣素组、XBPF组和顺铂组,干预15d后,观察抑瘤率,运用逆转录聚合酶链式反应(RT-PCR)法检测小鼠脾脏细胞中caspase-3基因的表达情况。结果:与模型组比较,XBPF组具有显著抑瘤作用(P<0.01),而且XBPF组的抑瘤作用强于多糖组和姜辣素组(P<0.05)。与模型组对比,XBPF组小鼠脾脏caspase-3基因的表达明显增多(P<0.01);XBPF组caspase-3基因的表达比多糖组和姜辣素组增多(P<0.05)。结论:XBPF对H22荷瘤小鼠肿瘤生长具有明显抑制作用,其机制可能与增强凋亡基因caspase-3表达有关。 Objective: To explore the antitumor effects of the tumor inhibitory of Xiaobanxia plus Fuling formula and its components and caspase-3 gene expression in splenic tissue of H22 tumor-bearing mice. Methods: 60 Mice were inoculated with H22 hepatoma cells to establish ascites H22 transplanted tumor model, then were divided into 6 groups: contral group, model group, Polysaccharide group, Gingerol group, Xiaobanxia Plus Fuling Decoction (XBPF) group and Cisplatin group. Suppression tumor rate was observed, and the mRNA expression variance of the caspase-3 in splenic tissues was detected by RT-PCR. Results: Compared with the model group, XBPF had obvious inhibition tumor cell proliferation in tumor-beating mice (P〈0.01), and significantly inhibited tumor growth than those of Polysaccharide group and Gingerol group (P〈0.05). The expression of caspase-3 mRNA in splenic tissues was significantly up-regulated compared with model group (P〈0.01). Furthermore, expression of caspase-3 gene in XBPF group was higher than those of Polysaccharide group and Gingerol group (P〈0.05). Conclusion: The anticancer effects of XBPF against H22 tumor-bearing mice are statistically significant, and its mechanism of action may be related to its modulation of caspase-3 expression.
