摘要
目的制备含有生殖支原体黏附蛋白(MgPa)模拟表位的多抗原肽(MAP),为研制基于模拟表位的MAP用于Mg的诊断与预防提供前期实验基础。方法以多聚赖氨酸为核心基质,采用Fmoc方法合成含有MgPa模拟表位的八分枝MAP,反向高效液相色谱(RP-HPLC)分析MAP的纯度,质谱分析法测定MAP的分子量以期对其进行鉴定。结果分别制备了含有MgPa 3个模拟表位的MAP,RP-HPLC分析的结果表明合成的3个MAP的纯度都在90%以上,质谱分析的结果说明所合成的MAP的分子量与预期的理论分子量相符。结论成功制备、纯化并鉴定了3个含有MgPa模拟表位的MAP。
Objective The multiple antigen peptides (MAP) containing the mimic epitope of the adhesion protein of Mycoplasma genitalium (MgPa) were prepared in order to provide the preliminary experimental basis for the development of mimotopes-baseded MAP used for the diagnosis and prevention of M.genitalium. Methods The eight branches MAPs containing the mimic epitope of MgPa were prepared by the Fmoc method using poly-lysine as the core matrix.The purity of MAPs were analyzed by reverse phase high performance liquid chromatography (RP-HPLC).The molecular weights of MAPs were characterizted by Mass Spectrometry. Results Three kinds of MAP containing the mimic epitopes of MgPa were successfully prepared.And the results of RP-HPLC showed that the purity of MAPs are above 90%.The molecular weight of MAPs are consistent with the expected theoretical molecular weight. Conclusion The 3 kinds of MAP,containing the mimic epitopes of MgPa,were successfully prepared,purified and identified.
