摘要
解析已育成的高产稳产油菜品种大地55的遗传基础,可以为后续油菜品种的培育提供借鉴和理论指导。本研究采用油菜60K SNP芯片对包括大地55亲本在内的5个保持系和7个恢复系间的遗传距离进行估算,发现大地55亲本间的遗传距离较大,为0.54,在所有35个组合中居第2位,是大地55杂交种能够产生强优势的遗传基础。用192对SSR引物对大地55的亲本1055A和R6进行扩增,并用杂种F1的DNA进行验证,筛选出4对可以对大地55进行纯度鉴定的引物(Ra2-G08,Ol11-H02,P052和P069)。采用Ra2-G08、Ol11-H02对大地55杂交种进行室内检测,纯度为94.3%,这批种子田间种植鉴定结果为95.1%,室内检测结果与大田种植鉴定的结果非常接近,说明用SSR标记可以对其种子纯度进行准确鉴定,从而为大地55在生产上安全应用提供保障。
Analysis of the genetic basis of high and stable yield varieties can provide theoretical guidance and reference for rapeseed breeding. The genetic distance between 5 maintainer lines and 7 restorer lines, including the parents of Dadi 55, was estimated by using 60K SNP (single nucleotide polymorphism) chip. Results showed that the genetic distance between the maintainer line and the restorer line of Dadi 55 was 0.54. It ranked the second among all 35 parental line combinations. The great genetic distance was one of the genetic bases for strong heterosis of this hybrid. The total of 192 pairs of SSR primers were used to amplify polymorphism bands between the genomic DNA of female parent 1055A and male parent Rr, and tested by DNA of F1 hybrid. Four pairs of SSR primers (Ra2-G08, O111-H02, P052 and P069) were screened for seed purity identification of Dadi 55. The seed purity identified by SSR primer combination Ra2-G08 and 111-H02 was 94.3%, this result was very closed to the field observation result (95.1%), indicating that seed purity of Dadi 55 can be identified accurately by SSR markers.
