HPLC法同时测定藏药铁棒锤中3种双酯型生物碱的含量

Simultaneous determination of three diester diterpenoid alkaloids inthe roots of Aconiti flavi et penduli by HPLC method

目的测定藏药铁棒锤中3种双酯型生物碱的含量。方法采用HPLC法对铁棒锤中双酯型生物碱进行含量测定。结果建立的高效液相色谱法专属性强；鸟头碱在0．06～0．65pg，次乌头碱在0·19～1．96μg，新乌头碱在0．22～2．25μg范围内呈良好的线性关系，r乌1=0.9996，r次=0.9994，r新=0.9992。平均回收率分别为96．9％、97．1％、96．3％，RSD分别为1．5％、1．8％、1．9％。结论建立的方法专属性强，灵敏度高，重现性好，可有效的控制铁棒锤的质量。

Objective To determine the content of diester diterpenoid alkaloids in Radix Aconiti flavi et penduli. Methods HPLC method was applied to determine the contents of diester diterpenoid alkaloids in the roots of Aconiti flavi et penduli. Results The study on the quality control showed that the new method by HPLC was good in sensitivity and specificity. The linear range of Aconitine, Hypaconitine, and Mesaconitine was 0.06-0.65 μg （r=0. 9996）, 0.19-1.96 μg （r=0. 9994）, and 0.22-2.25 μg （r=0. 9992） respectively. The average recovery （RSD） value of Aconitine, Hypaconitine, and Mesaconitine was 96.9% （1.5%）, 97.1% （1.8%）, and 96.3% （1.9%） respectively. Conclusion The method of identification and quantification is simple, reliable, and reproducible, and it can be used effectively for quality control of the roots of Aconiti flavi et penduli.
