摘要
内毒素刺激小鼠腹腔巨噬细胞和牛主动脉内皮细胞合成前列腺素I2、F2α、E2和血栓素A2。这种刺激作用有剂量和时间依赖性。对于预行色[^3H]花生四烯酸标记的内皮细胞,内毒素增加放射性花生四烯酸的释放和前列腺素I2的合成,多粘菌素B,放线菌酮和地塞米松抑制内素素对前列腺素合成的刺激作用。结果提示内毒素可能通过影响花生四烯酸由细胞脂质的释放过程刺激前列腺素合成。
The endotoxin stimulated immunoreactive PGI_2, PGF_(2α), PGE_2 and TXA_2 synthesis by mice resident peritoneal macrophages and bovine aorta endothelial cells. The stimulating effect depends on dose and time. The endotoxin also stimulated the release of [~3H] PGI_2 and [~3H] arachidonic acid from [~3H] araehidonic acid prelabelled endothelial cells. The endotoxin-stimulated prostaglandin production was inhibited by polymyxin B, cycloheximide and the antiinflammatory steroid, dexamethasone. Endotoxin might stimulate arachidonic acid metabolism by regulating liberation of esterified arachidonic acid from cellular lipids.
