利用组织芯片研究p21^(WAF1/CIP1)和Rb基因在肝细胞肝癌中的表达 被引量:6

摘要 目的:利用组织芯片研究肝细胞肝癌(hepatocelluarcarcinoma,HCC)中p21WAF1/CIP1简称p21、Rb蛋白的表达及两者的关系。方法:建立含40例HCC肿瘤组织及其相应癌旁、远癌正常组织共120个微组织的组织芯片,应用免疫组织化学技术(SP法)检测HCC肿瘤、癌旁、远癌正常组织中p21、Rb蛋白的表达情况。结果:p21蛋白缺失率在HCC肿瘤组织中为27.5%,显著高于癌旁及远癌正常肝组织(P<0.05),并与HCC的组织学分级相关(P<0.05);Rb蛋白缺失率在HCC肿瘤组织中为42.5%,显著高于癌旁和远癌正常肝组织P<0.05,但与HCC肿瘤的大小和组织学分级均无统计学相关性(P>0.05)。HCC肿瘤组织中p21与Rb的表达呈负相关P<0.05。结论:p21、Rb蛋白表达缺失可能与HCC的发生有密切联系,p21与Rb基因在HCC中可能通过负反馈机制相互调节。组织芯片是可高效进行肿瘤分子病理研究的技术平台。 Ob jective:To study the expression of p21 WAF1/CIP1 ,Rb proteins in hepatocellular carc inoma (HCC)and their relationship by tissue chip.Methods :A tissue chip which contained tumor t issues,correspondi ng adjacent liv er tissues and normal liver tissues of 40cases of HCC was cons tructed.The ex pression of p21and Rb proteins in tum or tissues,correspondin g adjacent liver tissu es and normal liver tissues of 40case s of HCC were stu died by SP immunohistochemical technique.Results:p21protein deletion rate in tumor tissues of HCC was 27.5%,which was high er than that in adjacent l iver tissues and normal liver tissu es(P<0.05).p21protein deletion cor related re markably with the pathological grad e of HCC(P<0.05).Rb prote in deletion rate in tumor tissues of HCC was 42.5%,which was also higher t han that in adjacent liver tissues and normal li ver tissues,Rb protein deletio n rate dosen' t correlated remarkably with tumor s ize or pathological grade of HCC(P>0.05).p21expression showed negative correlation with Rb expressi on(P<0.05).Conclusion:p21and Rb protein deletion may play a n importa nt role in tumorigenesis of H CC.There may be a negative feedback pathway betw een p 21and Rb in HCC.Tissue chip is an effective high-throughpu t technique platform for the study of tumor molecular pathology.
