摘要
目的:探讨黄芩、白术对小鼠胚胎着床的影响及其调控的糖分子机制。方法:将120只受孕雌鼠随机分为正常对照、障碍模型及中药治疗3组各40只。障碍模型组和中药治疗组小鼠经米非司酮诱导建立稳定的胚胎着床障碍模型,中药治疗组从妊娠第1天起每天中药灌胃,对照组和障碍模型组每日用等量生理盐水代替。孕5天取子宫内膜通过RT-PCR、Western blot检测着床相关寡糖合成关键酶的表达变化,孕8天观察胚胎着床数变化。结果:对照组平均胚胎着床数23.32±3.68,障碍模型组平均胚胎着床数3.00±2.00较对照组明显减少;中药治疗组平均胚胎着床数15.6±3.60较障碍组明显升高。RT-PCR及Western blot检测显示中药黄芩、白术治疗后岩藻糖基转移酶Ⅳ(FUT4)、Ⅶ(FUT7)表达明显高于障碍模型组。结论:黄芩、白术可通过调节着床相关寡糖合成关键酶FUT4、FUT7的表达来促进胚胎着床。
Objective: To explore the effects of Baikal Skullcap Root and Largehead Atractylodes Rhizome on embryo implantation in mice and the saccharide ,nolecular mechanism. Methods: A total of 120 pregnant mice were randomly divided into control group, model group and treatment group, 40 mice in each group. Stable embryo implantation dysfunction model was established by mifepristone in model group and treatment group, the mice in treatment group were treated with intragastric administration of Baikal Skullcap Root and Largehead Atractylodes Rhizome from the first day of pregnancy, while the mice in control group and model group were treated with intragastric administration of normal saline of the same dose from the first day of pregnancy. Endometrial tissue specimens were obtained on the fifth day of pregnancy, then RT - PCR and Western blot were used to detect the changes of expressions of embryo implantation - related key en- zymes of oligosaccharide synthesis ; the changes of numbers of embryo implantation on the eighth day of pregnancy were observed. Results : The average implantation number in model group was ( 3.00 ±2. 00), which was significantly lower than that in control group ( 23.32 ±3.68) ; the average implantation number in treatment group was (15.6 ±3.60), which was significantly higher than that in model group. RT - PCR and Western blot showed that the expression levels of FUT4 and FUT7 in treatment group were significantly higiler than those in model group. Conclusion : Baikal Skullcap Root and Largehead Atractylodes Rhizome can promote embryo implantation by regulating embryo implantation - related key enzymes ( FUT4 and FUT7) of oligosaccharide synthesis.
