二仙汤、二至丸和地黄煎对小鼠脾细胞和卵巢颗粒细胞的调节作用 被引量:9

摘要 目的探讨二仙汤、二至丸和地黄煎对小鼠脾细胞和卵巢颗粒细胞的调节作用。方法取小鼠脾脏和卵巢分别制备脾单个核细胞和卵巢颗粒细胞,于24孔板中培养,分别加入二仙汤、二至丸以及地黄煎的水提物和醇提物,以香菇多糖水溶剂为阳性对照药物,运用MTT法观察细胞增殖情况;酶联免疫吸附法(ELISA)测定细胞上清液中IL-1、IFN-γ及E2的浓度。结果二仙汤、二至丸、地黄煎各组对脾单个核细胞及卵巢颗粒细胞均具有较好的增殖作用(P<0.05,P<0.01);三方均可提高小鼠脾单个核细胞培养上清液中IL-1、IFN-γ的含量(P<0.05,P<0.01),但与阳性对照组相比,三方均无差异(P>0.05);三方均可提高小鼠卵巢颗粒细胞上清液中的E2水平(P<0.01,P<0.05);与阳性对照组相比,三方均无差异(P>0.05)。结论二仙汤、二至丸、地黄煎对小鼠脾细胞和卵巢颗粒细胞均有较好的调节作用。 Objective To explore the regulatory effects of "Erxian Decoction", "Erzhi Pills" and "Dihuang Decoction" on spleen cells and ovarian granulosa cells in mice. Methods Removed the mice spleen and ovaries, respectively made into splenic mononuelear cells and ovarian granulosa cells and cultivate in 24 orifices. Added water extract and alcohol extract of "Erxian Decoction", "Erzhi Pills" and "Dihuang Decoction" respectively. Lentinan water solvent as positive control drug. MTT method and enzyme linked immunosorbent assay were used to observe cell proliferation and measure the concentration of IL-1 and IFN-γas well as E2 in the supernatant fluid of cells. Results " Erxian Decoction", " Erzhi Pills" and " Dihuang Decoction" had good proliferation effects on splenic mononuclear cells and ovarian granulosa cells ( P 〈 0.05, P 〈 0.01 ) ; three prescriptions can improve the concentration of IFN-γ and IL- 1 in the supernatant fluid of splenic mononuclear cells in mice ( P 〈 0.01 or P 〈 0.05 ), but compared with the positive control group, these three prescriptions had no significant difference ( P 〉 0.05 ) ; three prescriptions can improve the concentration of E2 in the supematant fluid of ovarian granulosa cells in mice ( P 〈 0.01 or P 〈 0.05 ) ; compared with positive control group, these three prescriptions had no sinifienat difference ( P 〉 0, 05 ). Conclusion "Erxian Decoction", "Erzhi Pills" and "Dihuang Decoction" all has better effects on regulating splenic mononuelear cells and ovarian granulosa cells.
