摘要
目的:探讨黄芩、白术对胚胎体外黏附的影响及其调控的糖分子机制。方法:培养人子宫内膜细胞(RL95-2)和人绒毛膜细胞(JAR),并经米非司酮诱导后建立着床障碍模型。通过黏附实验观察不同浓度黄芩、白术水煎液对胚胎黏附率的影响,采用RT-PCR、Western blotting检测着床相关寡糖合成关键酶的表达变化。结果:黄芩、白术50 mg/L治疗组胚胎黏附率较模型组显著升高(P<0.05),着床相关的岩藻糖基转移酶Ⅳ、Ⅶ(FUT4、FUT7)的m RNA及蛋白表达显著增加(P<0.001)。结论:中药黄芩、白术可通过上调FUT4、FUT7的表达促进胚胎体外黏附。
Objective: To explore the effect of Baikal Skullcap root and LargeheadAtractylodes Rhi- zome on the adhesion rate and the saccharide molecular mechanism of embryo implantation dysfunctional model in vitro. Methods: The implantation model system in vitro of the human endometrium cells (RL95-2) and human embryo cells (JAR) was set up. And then the implantation dysfunctional model was established by mifepristone. The embryo adhesion rate was observed by adhesion experiment and the expression of implantation-related oligosaccharide synthesis key enzyme fucosyltransferases Ⅳ (FUT4), fucosyltransferases Ⅶ (FUT7) was detected by RT-PCR and Western blotting. Results: The expression level of the key enzyme FUT4, FUT7 was increased significantly after being dealt with Baikal Skullcap root and LargeheadAtractylodes Rhizome (50 mg/L). Consis- tent with the elevated expression of FUT4, FUT7, embryo adhesion rate was significantly increased in Baikal Skullcap root and Largehead Atractylodes Rhizome treatment group (P〈0.05) compared with the implantation dysfunction group. Conclusion: Baikal Skullcap root and Largehead Atractylodes Rhizome can play a crucial role in embryo adhesion by up-regulating the expression of the key enzyme FUT4, FUT7, which was involved in the synthesis of implantation-related oligosaccharides.
