MEKK1在B6-Co小鼠胚胎发育过程中的表达

摘要 为了检测MEKK1在B6-Co小鼠胚胎眼睑发育关键时期的表达,探讨其在该突变系小鼠EOB(出生时眼睛睁开)表型形成过程中的作用,试验剖取E16.5和E18.5小鼠,剪取整个头部,用4%多聚甲醛固定脱水,制作冰冻切片,通过免疫荧光技术检测MEKK1在小鼠胚胎眼睑部位的表达;剪取胎鼠皮肤,提取蛋白,以GAPDH为内参,通过Western-blot检测MEKK1在B6-Co小鼠胚胎中的表达情况。结果表明:与B6小鼠相比较,MEKK1在B6-Co小鼠胚胎眼睑发育关键时期的表达差异不显著;但游离C端在E16.5小鼠的表达明显要高于E18.5的表达,差异显著。说明MEKK1对促进小鼠眼睑融合起到了重要作用。 C57 BL/6 - Corneal opacity ( B6 - Co) mouse ; eyelids open at birth (EOB) ; eyelid development ; MEKK1 ; expression levels Abstract:To detect the MEKK1 expression during a critical period of eyelid development of the C57BL/6 -Corneal opacity (B6 -Co) mouse embryo, and explore its function in the formation of eyelids open at birth (EOB) phenotype in mice from the mutant line. The fetal mice at 16. 5 and 18.5 days of embryonic period were dissected, the entire heads were clipped and dehydrated with 4% paraformaldehyde fixation to prepare frozen sections, and then MEKK1 expression in mouse embryonic eyelids was detected using immunofluorescence technique. The skins of fetal mice were clipped to extract protein, and then MEKKI expression in the B6 -Co mouse embryo was detected by Western -blot, taking GAPDH as an internal reference control. The result showed that there was no significant difference on MEKK1 expression in the B6 - Co mice compared with the B6 mice, but the expression of free C - terminal domain of MEKK1 at 16.5 days of embryonic period was significantly higher (P 〈0.05 ) than that at 18.5 days of embryonic period. The results indicate that MEKK1 played an important role in the promotion of eyelid fusion in mice.
