Rho/ROCK信号通路对在异位子宫内膜间质细胞NF-κB、MMP-9和TIMP-1表达的影响 被引量:7

摘要 目的探讨Rho/ROCK信号通路激活后,在异位子宫内膜间质细胞中NF-κB、MMP-9和TIMP-1表达的变化情况及可能的作用机制,为临床提供新的治疗靶点。方法取90例卵巢处子宫内膜在异位病灶以及90例正常子宫内膜,分离、纯化间质细胞,建立体外间质细胞培养模型。酶免疫吸附的方法分析ROCKⅠ激酶活性。QPCR法检测NF-κB、MMP-9和TIMP-1的转录水平,免疫印迹(Western blot)法检测NF-κB、MMP-9和TIMP-1的蛋白表达水平。结果在异位子宫内膜间质细胞多呈梭形或者多角形,贴壁和增殖较快;在异位子宫内膜间质细胞的ROCKⅠ激酶活性和转录水平显著高于正常子宫内膜(P<0.01);在异位子宫内膜间质细胞的NF-κB、MMP-9转录水平和蛋白表达水平显著高于正常子宫内膜(P<0.01),而TIMP-1的转录水平和蛋白表达水平显著低于正常子宫内膜(P<0.01)。结论在异位子宫内膜间质细胞中Rho/ROCK信号通路处于激活状态,Rho/ROCK信号通路激活以后,在异位子宫内膜间质细胞NF-κB、MMP-9的表达均升高,TIMP-1的表达降低。其机制可能与该通路激活后引起下游基因表达变化有关。
