摘要
以美国红栌无菌苗为材料,建立基于植物微扩繁器的液体快繁体系。结果表明,以0.8 mg·L-1 6-BA、0.4 mg·L-1 NAA和20 g·L-1蔗糖的MS为液体培养基,芽增殖培养的浸没频率为12次·d-1,浸没时间为5 min·次-1,增殖倍数达6.0;以1.0 mg·L-1IBA和20 g·L-1蔗糖的1/2MS为液体培养基,生根培养的浸没频率为6次·d-1,浸没时间为5 min·次-1,生根率可达90%以上。在液体培养体系中,小植株生长迅速,健壮,繁殖周期比固体培养的缩短2周。
The liquid culture system of rapid propagation of Cotinus coggygria‘Royal purple’ was established based on plant micropropagation bioreactor, in which the tissue culture plantlets were taken as material. The re-sults showed that with the immersion frequency of 12 times per day for 5 mins each time, multiplication rate of shoots reached 6.0 in MS liquid medium supplemented with 0.8 mg·L-1 6-BA, 0.2 mg·L-1 NAA, and 20 g·L-1 sucrose. With the immersion frequency of 6 times per day for 5 mins each time, the rooting percentage of the adventitious shoots was above 90%in 1/2MS liquid medium supplemented with 1.0 mg·L-1 IBA and 20 g·L–1 sucrose. In vitro plantlets grew fast and strong, and the plant regeneration cycle was shortened by two weeks in the liquid culture system than that in solid culture.
