摘要
目的探讨红藻氨酸(KA)致癫模型中大鼠海马基质相互作用分子1(STIM1)钙释放激活Ca2+(CRAC)关键蛋白的表达及意义。方法选取2个月的雄性Wistar大鼠,将其完全随机分为模型组和对照组。模型组大鼠海马注射1.5μl(100 ng/μl)KA,分别于术后6、12、24 h取标本备测。分别使用Western Blot和免疫组化法测定STIM1含量和STIM1蛋白动态表达变化。结果与对照组相比,模型组海马组织内STIM1的表达水平随时间而升高,24 h达到高峰;STIM1蛋白表达也表现出同样的动态升高趋势;术后6~24 h模型组海马免疫组织化学法检测显示,模型组STIM1表达明显增加,并且随时间表达逐渐递增。结论在颞叶癫模型中,STIM1表达水平呈现逐渐增加趋势,提示STIM1表达增加可能在癫的发生机制中发挥着重要作用。
To investigate the kainic acid(KA)in hippocampus of rats with epilepsy induced by matrixmodel of molecular interaction in l(STIM1)of calcium release activated Ca2+(CRAC)expression and significance of key protein.Methods Male Wistar rats of 2 months,it will be randomly divided into model group and control group. Hippocampal injection in the rats of model group 1.5 μl(100 ng/L)KA,respectively.After 6,12,24 h specimens were detected.Respectively using Western Blot and immunohistochemical determination of STIM1 and STIM1 protein expression method of dynamic change.Results Compared with the control group,the expression level of hippocampus in STIM1 model group increased with time,peaking at 24 h;the expression of STIM1 protein also showed the same dynamic rising trend;display detection of 6-24 h model group hippocampus immunohistochemistry after operation,the model group,the expression of STIM1 increased obviously, and the expression increased gradually with time.Conclusion In a model of temporal lobe epilepsy, the expression level of STIM1 showed a gradual increasing trend,suggesting that increased expression of STIM1 may play an important role in the pathogenesis of epilepsy.
