模拟失重及再超重对猕猴腮腺组织热休克蛋白HSP70表达的影响 被引量:3

Effects of Simulated Weightlessness and High + Gx Exposure on Expression of Heat Shock Protein( HSP70) in Parotid Gland of Rhesus Macaque

摘要 目的探讨模拟失重30 d及再超重对猕猴腮腺组织中热休克蛋白HSP70表达的影响。方法选用雄性猕猴23只,随机分为对照组(A组),失重(B组),超重+13 Gx/230s(C组),失重后再超重(D组,依据不同的+Gx强度和时间分为4个亚组:分别是+11 Gx/270s D1组、+13 Gx/230s D2组、+15 Gx/200s D3组、+13 Gx/230s后恢复9 d D4组)。通过HE染色观察腮腺组织形态学的变化,通过免疫组织化学染色及Western blot技术检测HSP70在腮腺中的表达。采用SPSS17.0对数据进行统计分析。结果对照组腮腺结构基本正常,实验组腮腺组织中可见不同程度的局灶性淋巴细胞浸润、腺泡细胞萎缩,导管扩张,血管扩张充血;对照组腮腺组织HSP70表达阴性,实验组腮腺组织HSP70表达阳性,随着+Gx强度的加大而表达增强的趋势。实验组(除D4组外)HSP70在蛋白水平的表达显著高于对照组(P<0.05)。结论模拟失重、超重环境可造成猕猴腮腺出现轻度病理变化,腮腺组织中HSP70表达显著增强。 Objective To explore the effects of simulated weightlessness and high + Gx exposure on heat shock protein HSP70 expression on rhesus macaque parotid. Methods Twenty-three healthy male rhesus macaques were randomly divided into four groups : control group ( group A, n = 3 ), 30 d simulated weightlessness group (group B, n = 3 ), + 13 Gx/230 s group ( group C, n = 3 ), high + Gx after 30 d weightlessness exposure group ( group D, n = 14). The D group was further divided into four subgroups according to the + Gx load : + 11Gx/270s (group D1, n = 3), + 13 Gx/230 s (group D2, n = 4), + 15Gx/200s (group D3, n =4), + 13 Gx/230 s and 9 d of recovery (group D4, n = 3 ). Histological changes of parotid gland were observed by HE staining, and the expression of heat shock protein (HSP) 70 was detected by immunohistochemistry (IHC) and Western blot. Results Histological examination revealed that the parotid glands in group A were in normal structure, while aeinar cells atrophy, lymphocytic infiltration, vascular dilation and congestion of blood vessels could be observed in the other experimental groups. IHC staining showed that weak expression of HSPT0 was detected in parotid of the control group, whereas strong expression of HSP70 were found in the pa- rotids of the experimental groups, which expressed mostly in the epithelial cells of duct. Western blot analysis showed that HSP70 expression in the experimental groups (except group D4) increased significantly as com- pared with that in control group ( P 〈 0.05 ). Conclusion High + Gx exposure after 30 d simulated weightless- ness can cause pathological changes and up-regulate HSP70 expression in parotid gland of rhesus macaque.
