摘要
目的观察右美托咪定对脓毒症大鼠不同时期炎症介质的作用及其对生存率的影响。方法将90只雄性Wistar大鼠随机(随机数字法)分为三组:正常对照组(C组),脓毒症模型组(s组),右美托咪定治疗组(D组)。正常对照组静脉注射等量的生理盐水,其余两组通过静注内毒素构建脓毒症模型,并给予相应不同的处理,按时间点取血,观察造模48h内各组大鼠的行为改变和病死率;应用酶联免疫吸附法检测血液中的早期炎症介质肿瘤坏死因子-α、白介素-6和晚期炎症介质高迁移率族蛋白B1的含量变化。结果三组间大鼠病死率存在显著差别,肿瘤坏死因子α、白介素6、高迁移率族蛋白B1血清水平存在显著差异。结论右美托咪定对脓毒症大鼠早期和晚期炎症介质均有明显的抗炎作用,并可降低脓毒症大鼠病死率。
Objective To investigate the effect of dexmedetomidine (DEX) on inflammatory mediators in different periods of sepsis and its impact on survival rate of rats with sepsis. Methods Ninety male Wistar rats were randomly divided into three groups: control group (group C), sepsis model group (group S), and DEX treatment group (group D). Lipopolysaccharide (LPS) was injected to the vein to reproduce a classic sepsis model, while in control group, equivalent volume of NS was used instead. Different treatments were given to the corresponding groups, and then venous blood was taken at given intervals. Behavioral changes and mortality rate of rats within 48 hours after experiment were recorded. The levels of the early inflammatory mediators such as tumor necrosis factor-α ( TNF-α), interleukin-6 ( IL-6 ) and the late inflammatory mediators such as high-mobility group boxl protein ( HMGB1 ) in blood serum were detected by ELISA. Results Among three groups, there were significant differences in mortality rate of rats, the serum level of TNF-α, IL-6 and HMGB1 ( all P 〈 0. 05 ). Conclusion DEX has a certain anti- inflammatory effects on early and late stages of sepsis in rats with sepsis, and can also reduce the mortality rate.
