摘要
目的观察地塞米松对哮喘小鼠活化T细胞核因子c1(NFATc1)表达及气道炎症的影响。方法建立哮喘小鼠模型并收集支气管肺泡灌洗液(BALF),酶联免疫吸附法(ELISA)测定BALF中细胞因子白介素-4(IL-4)、白介素-5(IL-5)及γ干扰素(IFN-γ)水平,免疫组织化学法测定气道周围淋巴细胞中NFATc1蛋白的表达。结果哮喘小鼠IL-4、IL-5水平及NFATc1蛋白表达较正常组升高,差异有统计学意义(P<0.05);地塞米松干预组IL-4、IL-5水平及NFATc1蛋白表达较哮喘组降低,差异有统计学意义(P<0.05)。结论地塞米松可抑制NFATc1的表达,降低细胞因子IL-4、IL-5水平,减轻哮喘气道炎症。
【Objective】 To investigate the effects of Dexamethasone on the expression of nuclear factor of activated T cells c1(NFATc1) and airway inflammation in asthma model of mice. 【Methods】 A mouse model of asthma was established and bronchoalveolar lavage fluid(BALF) was collected. The levels of IL-4, IL-5and IFN-γ in BALF were measured by ELISA. The expression of NFATc1 in the airway peripheral lymphocytes was observed by immunohistochemistry.【Results】 There were significant increases in the levels of IL-4 and IL-5 and the expression of NFATc1 in the asthmatic group compared to the control group(P 〈0.05). Compared with those of the asthmatic group, the levels of IL-4 and IL-5 and the expression of NFATc1 in the Dexamethasone group were significantly decreased(P 〈0.05). 【Conclusion】 Dexamethasone can inhibit the expression of NFATc1, reduce the levels of IL-4 and IL-5 and relieve the airway inflammation.
