摘要
目的在大鼠糖尿病模型下,评价缝隙连接蛋白43(Cx43)的表达和硫化氢(H_2S)对该蛋白的干预作用。方法将大鼠随机分为4组:PBS处理正常大鼠组(PBS-N)、H_2S处理正常大鼠组(H_2S-N)、H_2S处理糖尿病大鼠组(H_2S-Dia)、PBS处理糖尿病大鼠组(PBS-Dia)。大鼠腹腔注射链脲佐菌素(STZ)建立糖尿病心肌模型;应用聚合酶链反应(PCR)、蛋白免疫印迹法(Western blot),分别检测Cx43在基因、蛋白上的表达。结果PBS-Dia组的Cx43 mRNA,Cx43蛋白表达较PBS-N组明显下降;H_2S-Dia组Cx43 mRNA,Cx43蛋白的表达较PBSDia组明显增加。结论糖尿病大鼠心肌组织Cx43在基因、蛋白水平上的表达均有不同程度下降,而H_2S可上调Cx43 mRNA,提高Cx43蛋白的表达水平,进而改善细胞间的传导。
Objective To explore the effect of hyperglycemia on connexin 43 protein expression with hydro-gen sulfideintervention in diabetic rats .Methods Rats were randomly divided into 4 groups:the PBS-N group,H2 S-N group,H2 S-Dia group and PBS-Dia group.Diabetic rats were established by intraperitoneal injections of streptozotocin .In RT-PCR method to detect the expression of Cx 43 mRNA in myocardial tissue .The expression of Cx 43 were analyzed by Western blot.Results Compared with PBS-N group,the expressions of Cx43 mRNA and Cx43 in PBS-Dia group were significantly decreased;Compared with PBS-Dia group,the expressions of Cx 43 mRNA and Cx43 in H2 S-Dia group were increased significantly .Conclusion H2 S can improve diabetic rats myocardial tissue electrophysiological remodeling by adjusting the expression of Cx 43 mRNA.
