桃红四物汤对光老化小鼠皮肤组织中Smad-3、TIMP-1、TIMP-2、AP-1蛋白表达水平的影响 被引量:9

Effect of Taohong Siwu Decoction on Expressions of Smad- 3,TIMP- 1,TIMP- 2 and AP- 1 Protein in Skin Tissue of Photoaging Mice

摘要 目的:研究桃红四物汤对光老化小鼠皮肤组织中Smad-3、TIMP-1、TIMP-2、AP-1蛋白含量的影响。方法:将30只小鼠随机分为6组,分别为中药低、中、高剂量组、空白对照组、模型对照组、阳性对照组(VE组)。对模型对照组、阳性对照组(VE组)和中药低、中、高剂量组小鼠进行皮肤光老化模型制备,空白对照组小鼠正常饲养。在造模前30 min,给空白对照组和模型对照组小鼠灌胃生理盐水,高、中、低剂量组小鼠灌胃相应剂量药液,阳性对照组(VE组)灌胃维生素E滴剂。造模完成后,采用Western-blot方法检测皮肤组织中MMP-1、MMP-3、MMP-12、Smad-2蛋白含量的表达水平。结果:与空白对照组比较,模型对照组小鼠皮肤组织中Smad-3、TIMP-1、TIMP-2蛋白含量的表达水平显著下降,差异具有统计学意义(P<0.05),AP-1蛋白表达水平显著上升(P<0.05);与模型对照组相比,中药低、中、高剂量组小鼠皮肤组织中Smad-3、TIMP-1、TIMP-2蛋白的表达水平显著上升(P<0.05),AP-1蛋白表达水平显著下降(P<0.05)。结论:桃红四物汤对皮肤光老化具有一定的保护作用。 Objective: To study the effect of Taohong Siwu Decoction on expressions of Smad- 3,TIMP- 1,TIMP- 2 and AP- 1 protein in skin tissue of photoaged mice. Methods: Thirty mice were divided into 6 groups( high dose group,moderate dose group and low dose group,blank control group,model control group and positive control group). Blank control group was normally fed and other groups were established photoaging model. Before 30 minutes of the preparation of photoaging model,we gave saline to blank control group and model control group,corresponding dose of liquid to high,medium and low dose groups,vitamin E drops to positive control group. The expression levels of expressions of Smad- 3,TIMP- 1,TIMP- 2 and AP- 1 protein in skin tissue were measured by Western- blot. Results: Compared with blank control group,expressions of Smad- 3,TIMP- 1 and TIMP- 2protein in skin tissue of model control group were significantly lower( P 〈0. 05) and expressions of AP- 1 protein in skin tissue of model control group were significantly higher( P 〈0. 05). Compared with model control group,expressions of Smad- 3,TIMP- 1 and TIMP- 2 protein in skin tissue of high,medium and low dose groups were significantly higher( P 〈0. 05) and expressions of AP- 1 protein in skin tissue of high,medium and low dose groups were significantly lower( P 〈0. 05). Conclusion: Taohong Siwu Decoction has a protective effect on the skin photoaging.
