糖络宁对STZ诱导DPN大鼠氧化应激的影响 被引量:10

The Effects of Tangluoning on Oxidative Stress of Diabetic Peripheral Neuropathy Rats

摘要 目的观察糖络宁对链脲佐菌素(STZ)诱导糖尿病周围神经病变(DPN)大鼠氧化应激的影响。方法SD大鼠60只,随机选取10只为正常对照组,其余50只大鼠一次性腹腔注射60 mg/kg STZ建立糖尿病大鼠模型,造模成功后随机分为模型对照组、α-硫辛酸组、糖络宁低剂量组和糖络宁高剂量组。药物干预12周后,检测大鼠血清中总抗氧化能力(T-AOC)及抗氧化酶如谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)等的活力,同时测定氧化应激产物如丙二醛(MDA)含量、活性氧自由基(ROS)和诱导型一氧化氮合酶(iNOS)的活力。结果与空白对照组比较,模型对照组T-AOC降低(P<0.01),抗氧化酶GSH-Px、SOD、CAT活力降低(P<0.01);氧化应激产物MDA、ROS含量升高(P<0.01),iNOS的活力升高(P<0.01)。与模型对照组比较,糖络宁低剂量组T-AOC升高(P<0.01),抗氧化酶GSH-Px、SOD、CAT活力升高(P<0.01);氧化应激产物MDA含量降低(P<0.01),ROS含量降低(P<0.05),iNOS的活力降低(P<0.01)。糖络宁高剂量组T-AOC的活力升高(P<0.05),CAT的活力升高(P<0.01);氧化应激产物MDA含量降低(P<0.01),ROS含量降低(P<0.05),iNOS的活力降低(P<0.01)。结论中药复方糖络宁可以明显提高具有抗氧化能力物质SOD、GPx、CAT含量和T-AOC的活力,清除对周围神经有害的氧化代谢产物及氧化应激活性物质MDA、ROS和iNOS的含量及活力,具有明显抗氧化应激能力,减轻氧化损伤,对糖尿病周围神经病变(DPN)有明显的保护作用。 Objective: To investigate the effects of Tangluoning on oxidative stress of diabetic peripheral neu- ropathy rats. Methods: 60 SD rats were randomly divided into the control group with 10 cases and the experi- mental group with 50 cases. Rats in the experimental group were intraperitoneal injected 60 mg/kg STZ to induce diabetic rat model. Rats with fasting blood glucose concentration greater than or equal to 16.7 mmol/L after 72 h were treated as diabetes and were randomly divided into the model group, ct-lipoic acid group (ALA),low dose of Tang Luo Ning group (LTLN) and high dose of Tang Luo Ning group (HTLN). After treated with Tang Luo Ning for 12 weeks,T-AOC,GSH-Px,SOD,CAT in serum were detected,and at the same time,the content of MDA, ROS and the activity of iNOS in serum were measured. Results: The data showed that compared with the control group,T-AOC, GSH-Px,SOD, CAT in the model group markably decreased (P〈 0.01 ),while the content of MDA and ROS and the activity of iNOS increased (P〈 0.01 ). Compared with the model group,T-AOC ,GSH-Px,SOD, CAT of LTLN treated group increased(P〈 0.01 ) ,while the content of MDA and ROS and the activity of iNOS de- creased(P〈 0.01 ). Compared with the model group T-AOC,CAT of HTLN treated group increased(P〈 0.05 or P〈 0.01) and the content of MDA and ROS and the activity of iNOS decreased (P〈 0.05 or P〈 0.01 ). Conclusion: Tangluoning can increase the vitality of GSH-Px, SOD, CAT and T-AOC, clear the content of MDA and ROS and decrease the vitality of iNOS with TLN prevented diabetic peripheral neuropathy by attenuating oxidative stressinjury.
