摘要
目的探讨c-Myc基因与慢性粒细胞白血病之间的关系。方法应用逆转录聚合酶链式扩增反应(RT-PCR)技术检测30例CML(其中慢性期11例,加速期8例,急变期11例)和10例对照(正常骨髓6例和非恶性血液病4例)骨髓单个核细胞中c-myc基因的表达,分析c-myc基因与CML患者不同临床阶段之间的关系。结果 c-myc基因在CML不同临床分期的表达水平均有所升高,CML慢性期患者和对照组之间的差异无统计学意义(P>0.05),CML加速期和急变期患者c-myc基因表达水平均明显高于慢性期和对照组(P<0.05),CML急变期患者较加速期患者c-myc基因表达水平更高,差异具有统计学意义(P<0.05)。结论 c-Myc基因在CML加速期和急变期的表达明显升高,提示c-myc基因高表达可能是CML病情进展与急变的机制之一。c-myc基因表达水平在CML患者病程监测中具有重要意义,甚至可能作为c-myc基因抑制剂应用于CML的依据。
Objective To investigate the relationship between c-Myc and chronic myelogenous leukemia. Methods Applica- tion of reverse transcription polymerase chain reaction(RT-PCR) technique for the detection of CML in 30 cases(including 11 cases of chronic phase,8 cases of accelerated phase, 11 cases of blastic phase) and 10 controls (6 cases of normal bone marrow and 4 cases 4 cases of non-hematologic malignancies) e-Myc gene expression in bone marrow mononuclear cells, analyze the relationship between different clinical stages the c-Myc gene in patients with CML. Results e-Myc gene expression levels in different clinical stages of CML were all increased,the difference between chronic phase CML patients and the control group was not statistically significant(P〉0.05) ,The patients of CML accelerated phase and blastie phase with c-Myc gene expression levels were significantly higher than those in chronic phase and the control group (P〈0.05),The patients of CML blastic phase with e-Myc gene expression levels higher than those in The patients of CML accelerated phase,the difference was statistically significant (P〈0.05). Conclusion c-Myc gene expression in CML accelerated phase and blastie phase significantly increased,suggesting that c-Mye gene expression may be one mechanism for the progression and blastic of CML. The expression level of c-Mye gene is important in the course of monitoring CML patients,Perhaps even as inhibitors of e-Mye gene is applied CML basis.
