摘要 目的探讨胃癌组织IFITM3基因表达对胃癌细胞侵袭和转移能力的影响。方法采用实时荧光定量PCR、免疫组织化学、Western blot等方法检测120例胃癌患者癌及癌旁组织IFITM3基因表达情况;运用Transwell侵袭实验和划痕实验检测转染shRNA-IFITM3质粒对胃癌细胞株(AGS和MKN45细胞)侵袭和转移能力的影响。同时观察裸鼠体内接种低表达的IFITM3胃癌AGS细胞后发生肺转移情况。结果胃癌组织IFITM3mRNA和蛋白表达水平均显著高于对应癌旁组织(P<0.05);AGS和MKN45细胞中IFITM3mRNA呈高表达。转染shRNA-IFITM3质粒后AGS和MKN45细胞侵袭能力均明显降低(P<0.05),AGS细胞迁移能力显著下降(P<0.05)。裸鼠体内接种低表达的IFITM3胃癌AGS细胞后肺转移率显著下降(P<0.05)。结论 IFITM3基因在胃癌组织呈高表达,IFITM3基因在胃癌的侵袭和转移过程中发挥重要作用。 Objective To investigate the effects of IFITM3 expression on the migration and invasion of gastric cancer cells.Methods The expression of IFITM3 in gastric cancer and adjacent tissues were detected by real-time PCR,immunohistochemistry and Western blot in 120 patients.The effects of shRNA-IFITM3 plasmid transfection on the migration and invasion of gastric cancer AGS and MKN45 cells were measured by transwell assay and scratch test.In addition,lung metastasis was observed after inoculation of AGS cells with low IFITM3 expression in nude mice.Results IFITM3 mRNA and protein expression in gastric cancer tissues was significantly higher than that in adjacent tissues(P〈0.05).IFITM3 was highly expressed in AGS and MKN45 cells.The invasion capacity of AGS and MKN45 cells and migration ability of AGS cells were significant reduced after shRNA-IFITM3 plasmid transfection(P〈0.05).Moreover,lung metastasis was significantly decreased after inoculation of AGS cells with low IFITM3 expression in nude mice(P〈0.05).Conclusion IFITM3 is highly expressed in gastric cancer and plays an important role in the invasion and migration of gastric cancer cells.
