摘要
目的研究六味五灵片(南五味子、女贞子、连翘等)抗肝纤维化作用的谱效关系,探索其药效物质基础。方法拉丁超立方法将六味五灵片中6味药材随机抽样成20个不同比例的组合,HPLC法获取其提取物的指纹图谱。以LX-2肝星状细胞为药效模型,MTT法测定各组的抗肝纤维化作用。利用主成分分析及相关分析法,对药效数据和指纹图谱信息进行处理。结果 20个不同比例组合提取物对LX-2细胞均有抑制作用,HPLC指纹图谱有23个共有峰,其中14(五味子醇甲)、20(五味子甲素)、17、19、21号峰与药效关系密切。结论六味五灵片的抗肝纤维化作用可能是多成分相互影响的结果。
AIM To study the spectrum-effect relationship of anti-hepatic fibrosis effect of Liuwei Wuling Tablets( Schisandra sphenanthera,Ligustrum lucidum,Forsythia suspensa,etc.) and to explore its effective substance basis. METHODS Latin hypercube sampling method was applied to randomly dividing six medicinal materials in Liuwei Wuling Tablets into twenty groups with different proportions. HPLC was used for obtaining the fingerprints of their extracts. With LX-2 hepatic stellate cells as efficacy model,the anti-hepatic fibrosis efficacies of various groups were determined by MTT method. Then the pharmacodynamic data and information of fingerprints were analyzed by principal component analysis( PCA) and bivariate correlation analysis. RESULTS The twenty groups exhibited inhibition effects on LX-2 cells. There were twenty-three common peaks in the HPLC fingerprints. Among them,Peak14( schisandrin),20( deoxyschizandrin),17,19 and 20 were closely related to the efficacy. CONCLUSION The anti-hepatic fibrosis effect of Liuwei Wuling Tablets may be the result of multi-component interaction.
