MCT1在鼻咽癌病人外周血MDSC中的表达及意义 被引量:2

摘要 目的检测单羧酸转运体1(MCT1)在鼻咽癌病人和正常人外周血髓系抑制性细胞(MDSC)中的表达并探讨其临床意义。方法随机收集经病理确诊的鼻咽癌病人和正常人外周血各10例,红细胞裂解液处理后,采用BD FACSAriaⅢ流式细胞仪分选出MDSC(CD11b^+CD33^+HLA-DR^-),q-PCR检测比较两组MCT1 mRNA水平的差异。结果鼻咽癌病人外周血中MDSC的比例(19.77±4.77)%显著高于正常人(0.84±0.18)%,P<0.001;且前者MCT1 mRNA水平(4.05±0.56)为后者(2.15±0.33)的1.88倍,差异有显著性意义(P<0.01)。结论鼻咽癌病人外周血中的MDSC可能是凭借其在数量上的优势及能量代谢上的特性,更好地发挥免疫抑制效应。 Objective To investigate MCT1 expression of MDSC in peripheral blood obtained from nasophal:cngeal carcino- ma (NPC) patients and healthy donors, and discuss its clinical significance. Methods Peripheral blood samples were ob- tained randomly from 10 NPC patients and 10 healthy donors, processed by RBC lysates and MDSC ( CD11b + CD33 + HLA - DR- ) within them were sorted by BD FACSAriam. qRT - PCR were adopt to determine the MCT1 mRNA levels, t - test was used to evaluate the significance between groups. Results The MDSC ratio of NPC patients ( 19.77 + 4.77 ) % was significant higher than that of healthy donors (0.84±0.18 ) % ( P 〈 0. 001 ). MCT1 mRNA level (4.05±0.56) of MDSC obtained from NPC patients was significantly higher than that of healthy donors (2.15 ±0.33 ) (P 〈 0.01 ). Conclusion MDSC within peripheral blood of NPC patients could play an immunosuppressive effect by its superiority in number and energy metabolism property.
