针刺调节大鼠在脑卒中痉挛状态下黑质纹状体内相关神经兴奋-抑制因子受体的研究 被引量:18

Study on acupuncture regulating the receptors of nerve excitatory-inhibitory factor in nigral striatum of rats with spasticity of cerebral apoplexy

摘要 目的:探究针刺对脑卒中痉挛状态(SCA)大鼠大脑黑质纹状体中神经兴奋-抑制因子(Glu-GABA)含量与表达的调节作用,从而论证针刺对SCA的治疗作用。方法:32只SD大鼠被随机分为针刺组、空白组、假手术组、模型组。运用线栓法制作大脑中动脉缺血模型,造模成功后第1天开始治疗,连续3d。以Zealonga评分评价模型和针刺疗效,首先通过酶联免疫法检测大鼠大脑黑质中Glu、GABA的含量,然后运用RT-PCR法检测黑质中mGluR1a、GABABR1 mRNA的表达。结果:治疗前,与空白组、假手术组比较,模型组、针刺组Zealonga评分显著升高(P<0.01);治疗后,与模型组比较,针刺组较评分显著降低(P<0.01)。与空白组、假手术组比较,模型组GABA含量显著降低而Glu含量显著升高(P<0.01,P<0.05);与模型组比较,针刺组GABA含量升高而Glu含量显著降低(P<0.01,P<0.05)。与空白组、假手术组比较,模型组GABABR1 mRNA的表达显著减少而mGluR1a mRNA表达显著增强(P<0.01);与模型组比较,针刺组GABABR1 mRNA表达增强而mGluR1a mRNA表达减弱(P<0.05)。结论:针刺可调节大脑黑质中Glu和GABA的表达使其逐渐接近正常水平,说明了针刺对SCA有良好的治疗效果。 Objective:To observe and analyze the effect of acupuncture on the quantity and expression of neuronal excitatory-inhibitory factor(Glu-GABA)in nigral striatum of rats with SCA through experimental research,so as to demonstrate the treatment effect of acupuncture on spasticity of cerebral apoplexy.Methods:Thirty-two SD rats were randomly divided into acupuncture group,control group,sham operation group and model group.The MCAO model was established by using suture embolization.The treatment began on the first day after successful modeling and lasted for 3 days.Zealonga score was used to evaluate the model and the effect of acupuncture.The contents of Glu and GABA in substantia nigra of rats were detected by enzyme-linked immunosorbent assay(ELISA),and the expressions of mGluR1 a and GABABR1 mRNA in substantia nigra were detected by RT-PCR.Results:Before treatment,compared with the control group and the sham operation group,the Zealonga score of model group and acupuncture group were significantly higher(P<0.01).After treatment,the score of acupuncture group was significantly lower than that of model group(P<0.01).Compared with control group and sham operation group,GABA content in model group decreased while Glu content increased(P<0.01,P<0.05).Compared with model group,GABA content increased in the acupuncture group while Glu content decreased significantly(P<0.01,P<0.05).Compared with the control group and sham operation group,the expression of mGluR1 mRNA increased significantly in model group,while the expression of GABABR1 mRNA decreased significantly(P<0.01).Compared with model group,mGluR1 mRNA expression decreased and GABABR1 mRNA expression enhanced in acupuncture group,the difference was statistically significant(P<0.05).Conclusion:Acupuncture can regulate Glu and GABA in nigral striatumand to the normal level gradually,which shows that acupuncture has a good therapeutic effect on relieving spasticity of cerebral apoplexy.
