胡椒碱对胰岛素抵抗综合征模型大鼠脂代谢及作用机制的研究 被引量:5

摘要 目的探讨胡椒碱(PIP)对胰岛素抵抗综合征(IRS)大鼠甘油三酯(TG)、游离脂肪酸(FFA)、肿瘤坏死因子(TNF-α)、瘦素(Leptin)的作用。方法采用高脂高糖高盐饲料喂养制备IRS大鼠模型。每4周观察各组空腹TG,第4周开始给药,连续给药8周后,分别采用GPO-PAP法、比色法检测TG和FFA水平,ELISA法检测TNF-α、Leptin水平。结果给药8周后,模型大鼠血清TG、FFA、TNF-α、Leptin水平明显升高,不同剂量PIP使TG、FFA、TNF-α、Leptin水平下降。结论 PIP明显改善IRS模型大鼠的脂代谢异常情况,能降低IRS模型大鼠TG和FFA水平,其作用机制可能与下调细胞因子TNF-α、Leptin有关。 Aim To investigate the effect of piperine( PIP) on triglycerides( TG),free fatty acid( FFA),tumor necrosis factor-α( TNF-α) and leptin of model rats with insulin resistance syndrome( IRS). Methods By feeding rats with high-fat,high-glucose and highsalt diet,IRS model rats were established. Triglycerides( TG) was observed dynamically every 4 weeks,treating on the 4th weeks and 8 weeks continuously.TG( method of GOD-POD),FFA( method of colorimetric),TNF-α and leptin( method of ELISA) were measured. Results After 8 weeks treatment,the level of TG,FFA,TNF-α and leptin in PIP group decreased obviously,while it increased in model group significantly. Conclusions PIP could reduce TG and FFA and improve abnormal lipid metabolism of model rats with IRS significantly. Perhaps the mechanism is related with the down-regulation of cytokine TNF-α and leptin.
