摘要
目的:探讨不同剂量的醋酸丙氨瑞林对人子宫内膜癌裸鼠皮下移植瘤生长的影响及TRF1的表达情况。方法:采用HEC-1-B细胞成功建立人子宫内膜癌裸鼠皮下移植瘤模型,选取34只雌性荷瘤裸鼠随机分为四组:对照组、低剂量醋酸丙氨瑞林组(20μg/kg)、中剂量醋酸丙氨瑞林组(40μg/kg)、高剂量醋酸丙氨瑞林组(80μg/kg)。观察裸鼠生长发育情况,每7天测量皮下移植瘤体积,4周后处死裸鼠,取出移植瘤,测量肿瘤体积,计算抑瘤率。移植瘤组织石蜡包埋行苏木素-伊红(HE)染色,镜下观察瘤细胞形态变化。免疫组织化学法检测各组瘤组织中端粒重复因子1(TRF1)的表达情况。结果:低、中、高醋酸亮丙瑞林组的抑瘤率逐渐增高,分别为15.03%、21.80%及39.98%,各组间两两比较差异有统计学意义(P<0.05)。HE染色后干预组瘤细胞排列较对照组稀疏,瘤细胞核深染、固缩。随着药物剂量增加,免疫组织化学检测到瘤组织中TRF1表达下调,各干预组与对照组相比,差异有统计学意义(P<0.05)。结论:醋酸丙氨瑞林对子宫内膜癌细胞株HEC-1-B裸鼠皮下移植瘤生长有抑制作用,可能与TRF1蛋白表达下调有关。
Objective: To explore the effect of intervention with Alarelin on telomeric repeat binding factor - 1 ( TRF1 ) ex- pressions in endometrial cancer and subcutaneously xenografted tumor in nude mice. Methods: Endometrial cancer cell line HEC - 1 - B cultured in vitro was inoculated into subcutaneous tissue of nude mice to establish xenograft tumor model. 3 4 female nude mice were randomly divided into control group ( normal saline ) , different dose group ( 2 0 μg / kg, 4 0 μg / kg, 8 0 μg/kg). During the period of treatment, our group members measured the size and volume of tumor every seven days. After four weeks, we were removed the xenograft tumor and calculate the volume of tumor. The changes of xenograft morphology were observed by light microscopy with Hematoxylin- easin(HE) staining, and the change of TRF1 (TRF1) expressions were esti- mated by immunohistochemieal technology. Resuhs: Alarelin inhibited the tumor growth by 15.03%, 21.80%, 39.98%, in difference alarelin groups, and the difference was significant between the groups ( P 〈 0.05 ). The hematoxylin - easin staining results showed significant changes. With the increase of alarelin, the expression of TRF1 is decreased. Conclusion :Alarelin have direct inhibitory effect on nude mice who bearing human endometrial carcinoma, which may be associated with the decreased expression of TRF1 protein.
