B7-H3和Smad7蛋白在食管鳞状细胞癌组织中的表达及临床意义

Expression of B7-H3 and Smad7 protein in esophageal squamous cell carcinoma tissue and the clinical significance

目的探讨B7-H3和Smad7蛋白在食管鳞状细胞癌(ESCC)组织中的表达水平及临床意义。方法收集82例ESCC组织标本及其癌旁正常组织石蜡标本,运用免疫组织化学方法检测B7-H3及Smad7蛋白的表达。采用实时荧光定量PCR(qRT-PCR)检测组织中B7-H3 mRNA的表达水平。结果 B7-H3在ESCC组织中的表达阳性率高于癌旁正常组织,Smad7蛋白表达阳性率低于癌旁正常组织(P<0.05)。ESCC组织中B7-H3和Smad7蛋白的表达呈负相关(rs=-0.294,P=0.005)。ESCC组织中B7-H3蛋白表达与临床分期、分化程度有关(P<0.05),Smad7蛋白表达与分化程度有关(P<0.05)。结论 ESCC组织中B7-H3蛋白表达上调,Smad7蛋白表达下调。B7-H3和Smad7蛋白的异常表达可能参与了ESCC的发生发展过程。

Objective To explore the expression of B7-H3 and Smad7 protein in esophageal squamous cell carcinoma( ESCC) tissue and the clinical significance. Methods Eighty-two cases of ESCC tissues and adjacent normal tissues were selected and the expression of B7-H3 and Smad7 protein were detected by immunohistochemistry. Quantitative real-time polymerase chain reaction( qRT-PCR) was used to detect the expression level of mRNA B7-H3 in ESCC tissues and adjacent normal tissues. Results Positive expression rate of B7-H3 protein in ESCC tissues was significantly higher than the adjacent normal tissues( P < 0. 05),and the positive expression rate of Smad7 protein was lower than the adjacent normal tissues( P < 0. 05). The expression of B7-H3 protein was negatively correlated with Smad7 protein in ESCC tissues( rs=-0. 294,P = 0. 005). Expression of B7-H3 protein was significantly related to clinic staging and differentiation grade( P < 0. 05),and the expression of Smad7 protein was significantly related to differentiation grade( P < 0. 05). Conclusion Expression of B7-H3 protein was up-regulated and expression of Smad7 protein was down-regulated in ESCC tissues. The abnormal expression of B7-H3 and Smad7 protein may be involved in the occurrence and development of ESCC.
