摘要
目的对15批不同等级的大海马进行红外光谱分析,研究红外指纹图谱在其品质分析中的应用,验证市场分级现状是否合理,为海马的品质鉴定提供快速有效的方法。方法利用傅里叶变换红外光谱法(FTIR)的一维光谱图谱的比较分析,结合粗脂肪与总酚含量测定。结果建立了不同等级海马的指纹特征,发现不同等级大海马的粗脂肪与总酚含量差异显著。劣质品海马的一维指纹图谱与正常品明显不同,相似度低;海马的粗脂肪与商品等级呈极显著正相关水平,为0.821;海马的总酚含量与商品等级呈显著正相关水平,为0.607;根据1 092.4及2 932.2cm^(-1)处的二阶导数值建立的判别函数,可用于判别海马的等级。结论红外指纹图谱专属性强、快速、稳定,可为海马的质量鉴别研究提供科学依据。
Objective Infrared fingerprints of fifteen batches of Hippocampus kuda were studied to verify if the status of the market classification was reasonable and it could be used for quality analysis.Methods Fourier transform infrared spectroscopy(FTIR)combined with one-dimensional spectroscopy,similarity comparison,and second derivative spectra were also used to find out the characteristics and differences of each sample.Then the crude fat and total phenol content were measured and compared.Results Characteristics and differences among fingerprints were found out.Meanwhile,crude fat and total phenol content of different grades were significantly different.One-dimensional fingerprints of the poor Hippocampus were easily identified from the normal ones,the similarity was low.Commercial grades were significantly correlated with the crude fat content and the total phenol content,which were0.821 and 0.607,respectively.Second derivative spectra at 1 092.4 and 2 932.2 cm^(-1) could be used to determine the grade of hippocampus.Conclusion FTIR was specific,rapid and stable.It could be used for the quality identification of Hippocampus.
