走马胎水提取物对肝癌的抑制作用及其机制 被引量:4

Inhibitory effect and mechanism of Ardisia gigantifolia water extract on liver cancer

摘要 目的探讨走马胎水提取物对肝癌的抑制作用及其可能机制。方法体外培养人肝癌HepG2和SK-hep1细胞,随机均分为走马胎水提取物组(A组)和阴性对照组(B组)。采用MTT法检测走马胎水提取物对肝癌细胞生长的抑制作用,倒置荧光显微镜下观察走马胎水提取物对细胞凋亡的影响,实时定量PCR检测细胞周期素依赖性激酶2(CDK2)、Caspase-3及上皮细胞钙黏蛋白(E-cadherin)mRNA表达。结果走马胎水提取物对HepG2和SK-hep1细胞均有明显抑制作用,IC50分别为(3.21±0.35)mg/ml和(2.17±0.10)mg/ml(P<0.05)。倒置荧光显微镜下观察发现走马胎水提取物能明显诱导两种肝癌细胞的凋亡。与B组相比,A组HepG2细胞中CDK2mRNA表达降低,而Caspase-3和E-cadherin mRNA表达增加(P<0.05或P<0.01)。结论走马胎水提取物可能通过下调CDK2表达、上调Caspase-3和E-cadherin表达,从而抑制肝癌的发生和发展。 Objective To investigate the inhibitory effect and underlying mechanism of Ardisia gigantifolia water extract on liver cancer. Methods Human liver cancer cell lines HepG2 and SK-hepl were cultured in vitro and randomly divided into two groups of A (treated with Ardisia gigantifolia water extract 2.84 mg/ml) and B(negative control). The inhibitory effect of Ardisia gigantifolia water extract on cell growth was determined by MTT assay. The cell apoptosis was observed under inverted fluorescence microscope. The mRNA expressions of eyclin-dependent kinase2(CDK2),Caspase-3 and E-cadherin were detected by qRT-PCR. Results The growth of HepG2 and SK-hepl cells was obviously inhibited by Ardisia gigantifolia water extract and the values of IC50 were (3.21 ±0.35) mg/ml and (2.17±0. 10) mg/ml, respectively (P〈0. 05). The apoptosis of HepG2 and SK-hepl cells was significantly induced by Ardisia gigantifolia water extract. Compared with group B, the mRNA expression of CDK2 was decreased, which of Caspase-3 and E-cadherin was increased in HepG2 cells in group A(P〈0. 05 or P〈0. 01). Conclusion Ardisia gigantifolia water extract can inhibit the development and progression of liver cancer probably via downregulating CDK2 expression and upregulating Caspase-3 and E-cadherin expressions.
