高效液相色谱法测定黄连上清丸中芦荟大黄素和大黄酸的含量 被引量:1

Simultaneous Quantification of Aloe Emodin and Rhein in Huanglian Shangqing Pills by High Performance Liquid Chromatography

摘要 目的建立黄连上清丸中芦荟大黄素和大黄酸含量的测定方法。方法采用高效液相色谱法测定黄连上清丸中芦荟大黄素和大黄酸的含量,色谱柱为Inertsil-ODS3-C18(4.6mm×150mm,5μm),流动相为甲醇(A)-0.2%磷酸溶液(B)(6535),检测波长为254nm,流速为1.0mL·min^(-1),柱温为25℃。结果芦荟大黄素在8.28~41.4μg·mL^(-1)与峰面积呈良好的线性关系(r=0.999 9),大黄酸在5.94~29.7μg·mL^(-1)峰面积呈良好的线性关系(r=0.999 8)。芦荟大黄素、大黄酸的平均加样回收率分别为100.50%、98.74%,RSD分别为2.03%、0.23%。结论建立的高效液相色谱法具有简单、灵敏、重复性好等优点,可作为黄连上清丸中芦荟大黄素和大黄酸含量的测定方法。 Objective To establish a HPLC method for the quantification of aloe emodin and rhein in Huanglian Shangqing pills. Methods HPLC method was used to determine the contents of aloe emodin and rhein in Huanglian Shangqing pills. The analysis was carried out on an Inertsil-ODS3-CiS column (4. 6 mmX150 m m,5 jLtm),with a mobile phase of methanol (A )-〇. 2 % phosphoric acid solution (B) (65 : 35). The flow rate, detection wavelength and column temperature were 1 .〇 mL ? min—1 ,254 nm and 25 °C , respectively. Results The linear calibration range was 8. 28-41. 4 fj.g ? mL_1 aloe emodin (r= 0. 999 9) ,and 5. 94-29. 7 ? mL_1for rhein (r = 0. 999 8).The average recovery and RSD were, respectively, 100. 50% and 2. 03% for aloe emodin, and 98. 74% and 0. 23% for rhein. Conclusion The HPLC method is simple,sensitive and reproducible for the determination of aloe emodin and rhein in Huanglian Shangqing pills.
