淫羊藿女贞子影响激素干预哮喘大鼠与炎性因子相关的pJAKs/pSTATs的研究 被引量:5

Effects of Epimeium and Fructus Ligustri lucidi on pJAKs/pSTATs associated with inflammatory factors in asthmatic rats treated with hormone

摘要 目的研究淫羊藿女贞子合地塞米松对哮喘大鼠肺组织中与炎性因子相关的JAKs及STATs蛋白磷酸化表达的影响。方法 SD雄性大鼠,随机分为7组,分别是正常对照组(正常组)、哮喘模型组(模型组)、地塞米松组(激素组)、淫羊藿女贞子煎剂组(煎剂组)、地塞米松合煎剂组(激素合煎剂组)、淫羊藿女贞子提取物组(提取物组)、地塞米松合提取物组(激素合提取物组)。卵蛋白(OVA)致敏,建立大鼠哮喘模型,腹腔注射地塞米松和/或灌胃淫羊藿女贞子煎剂或提取物,免疫荧光法检测肺组织JAKs(JAK3、TYK2)及STATs(STAT3、STAT5、STAT6)蛋白的磷酸化表达。结果与正常组相比,模型组大鼠肺组织p JAK3、p TYK2、p STAT3、p STAT5、p STAT6的蛋白表达均显著升高(均P<0.01);与模型组比较,各给药组p JAK3、p TYK2、p STAT3、p STAT5、p STAT6的蛋白表达均显著降低(均P<0.01);与单用激素比较,激素合煎剂或提取物组均能进一步显著降低p JAK3、p TYK2、p STAT3、p STAT6的蛋白表达。结论调节哮喘大鼠肺组织中与炎性因子相关的JAKs/STATs可能是淫羊藿女贞子煎剂及提取物协同激素治疗哮喘的作用机制之一。 Objective To study the effects of the decoction and extracts of Epimeium and Fructus Ligustri lucidi(EF)on JAKs/STATs protein phosphorylation which connected with inflammatory factors in lung tissues of asthmatic rats induced by dexamethasone.Methods The SD rats were randomly divided into seven groups,including normal control group(normal group),asthma model group(model group),dexamethasone group(hormone group),EF decoction group(decoction group),EF decoction plus dexamethasone group(hormone plus decoction group),EF extracts group(extract group),and EF extracts plus dexamethasone group(hormone plus extract group).Asthmatic rat model was induced by the injection and atomization inhalation of OVA.Intraperitoneal injection of dexamethasone and/or gavage of EF decoction or EF extracts.The phosphorylation protein expressions of JAKs(including JAK3、TYK2)and STATs(including STAT3、STAT5、STAT6)of lung tissues were detected by immunofluorescence method.Results Compared with the normal group,the protein expression of pJAK3,pTYK2,pSTAT3,pSTAT5and pSTAT6in lung tissue in the model group were significantly increased(P<0.01);compared with the model group,the protein expression of pJAK3,pTYK2,pSTAT3,pSTAT5and pSTAT6in treatmet group were significantly degraded(P<0.01);compared with dexamethasone group,the protein expression of pJAK1,pJAK2,pSTAT1,pSTAT2,pSTAT4in the EF decoction plus dexamethasone group or EF extracts plus dexamethasone group were significantly decreased(P<0.01).Conclusion The mechanisms of Epimeium and Fructus Ligustri lucidi decoction and its extract combined with corticosteroids in the treatment of asthma may be to regulate the JAKs/STATs related to inflammatory factors in the lung tissue of asthmatic rats.
