规模化猪场伪狂犬病净化方案应用效果的研究

摘要 猪伪狂犬病被世界动物卫生组织(OIE)列为B类动物疫病,在我国一些大型规模化猪场普遍存在,直接或间接影响我国养猪业的健康发展,该病已被列入《国家中长期动物疫病防治规划》(2012-2015年)重点优先防制和净化的疫病。本研究是在应用猪伪狂犬病gE/gB-ELISA监测方法(GB/T 18641-2002)进行规模化猪场猪伪狂犬病抗体监测区分PRV野毒与疫苗毒感染的基础上,配合猪场从PRV病原学检测区分PRV野毒与疫苗毒的多重PCR(mPCR)方法和生物安全措施的建立,对贵州某规模化猪场伪狂犬病进行净化方案应用效果进行研究。先后开展了对安顺某规模化猪场血液进行了4次PRV-gB/gE ELISA抗体监测及PRV抗原检测,共监测样品为1 837份,检测结果表明,安顺某规模化猪场在开展伪狂犬病净化工作之后猪PRV-gB抗体水平均达到90%,PRV-gE抗体阳性率及PRV抗原阳性率均降为0,表明所开展的净化方案对规模化猪场猪伪狂犬病的净化工作有明显的效果,该研究可为规模化猪场伪狂犬病的净化提供参考。 Pseudorabies is classified as a B class animal disease by Office International Des Epizooties(OIE). It is common in some large-scale pig farms in China. It directly or indirectly affect the healthy development of pig industry in China. The disease has been included in the national long-term animal disease control program(2012-2015 years). This study was performed to to monitor pseudorabies antibody in large-scale pig farms using the gE/gB-ELISA surveillance method(GB/T 18641-2002), and to distinguish PRV from wild virus and vaccine virus. The multiplex PCR(mPCR) method and biosafety measures of PRV wild virus and vaccine toxicity were established from the PRV etiology tests. The elimination of pseudorabies in a large-scale pig farm in Guizhou was studied. 1837 blood samples from different stage pigs(boar, reserve pigs, nursing pigs, and fattening pigs) were collected to monitor PRV-gB/gE antibody by PRV-gB/gE ELISA and PRV antigens by PCR 4 times in Anshun, The results showed that after the purification of pseudorabies in anshun, the pig PRV-gB antibody level reached 90%, and the positive rate of PRV-gE antibody and PRV antigen were reduced to 0, indicating that the scheme had obvious effect on the purification of scale swine pseudorabies.
