摘要
目的:探讨Toll样受体4(TLR4)/髓样分化蛋白88(MyD88)与早期自然流产的相关性.方法:选择30例正常妊娠妇女作为对照组,采用免疫组织化学显色对30例早期自然流产患者(流产组)母-胎界面的蜕膜和绒毛组织中TLR4、MyD88蛋白表达进行定位观察,采用RT-PCR技术对其TLR4和MyD88 mRNA表达进行半定量分析.结果:TLR4和MyD88蛋白在流产组和对照组的绒毛及蜕膜组织中均有表达,定位表达于绒毛滋养层细胞、蜕膜细胞的细胞质内,阳性反应强弱不等.RT-PCR半定量分析结果显示,在绒毛组织中,流产组TLR4 mRNA、MyD88 mRNA的表达显著高于对照组;在蜕膜组织中,流产组TLR4 mRNA、MyD88 mRNA的表达显著高于对照组.流产组绒毛和蜕膜组织中TLR4与MyD88的表达均呈正相关.结论:TLR4和MyD88可能参与早期自然流产的病理过程,且TLR4可能通过MyD88信号途径参与早期自然流产的发生.
Objective :To explore the correlation between Toll-like receptor 4(TLR4)/myeloid differentiation protein 88(MyD88) and early spontaneous abortion. Methods :Immunohistochemical staining was used to localize the expression of TLR4 and MyD88 proteins in decidua and villi of 30 patients with early spontaneous abortion (abortion group). The expression of TLR4 and MyD88 mRNA was semi-quantitatively analyzed by RT-PCR. Thirty normal early pregnant women were selected as controls. Results :TLR4 and MyD88 proteins were expressed in the villi and decidua of both abortion and control groups,and were localized in cytoplasm of trophoblast cells and decidual cells. The results of RT-PCR semi-quantitative analysis showed that the expression of TLR4 mRNA and MyD88 mRNA in the villi in the abortion group was significantly higher than that in the control group. The expression of TLR4 mRNA and MyD88 mRNA in the decidua in the abortion group was significantly higher than that in the control group. The expression of TLR4 was positively correlated with MyD88 in the villi and decidua of abortion group. Conclusion : TLR4 and MyD88 may participate in the pathological process of early spontaneous abortion,and TLR4 may play important roles in the occurrence of early spontaneous abortion through MyD88 signaling pathway.
