CD3/CD8设门在FCM检测Th1/Th2亚型中的应用

Application of CD3/CD8 gating to examination of Th1/Th2 cells in flow cytometry

目的 探讨流式细胞术准确、灵敏检测Th1/Th2细胞的新方法。方法 采用荧光mAbCD3和CD8设门 ,准确找到CD4 + T细胞群 ,然后进行Th1/Th2细胞分析 ,并与单用CD4设门进行比较。结果 以CD3/CD8设门较单用CD4设门 ,对靶细胞的定位较为准确 ,测量所获图形美观、清楚并有规律。结论 以CD3/CD8设门 ,可提高检测Th1/Th2细胞的准确性 ,同时也可应用于Tc1/Tc2细胞的检测分析 。

Aim To find a new approach for more accurate and sensitive detection of Th1/Th2 cells by flow cytometry. Methods Fluorescent mAb CD3 and CD8 gating were used to accurately detect CD4 + T cells. Th1/Th2 cells were then analyzed and compared with the result obtained with CD4 gating alone. Results In localizing target cells, the CD3/CD8 gating technique was better than that of using CD4 alone in the accuracy, clarity and regularity of diagrams. Conclusion CD3/CD8 gating can improve the accuracy of detecting Th1/Th2 cells by flow cytometry and can also be applied to the examination of Tc1/Tc2 cells. Therefore, it is a rather ideal and practical means of examining samples in clinical and basic immunological studies.
