摘要
目的识别胆管癌致病的关键基因及信号通路,为寻找治疗靶点提供数据支持。方法利用GEO数据库的两个基因芯片数据集GSE26566和GSE76297进行差异基因的筛选,并通过DAVID数据库对其进行功能富集分析。采用STRING数据库构建了基因编码蛋白之间蛋白质-蛋白质相互作用网络,随后导入Cytoscape软件进行可视化分析。通过TCGA和The Human Protein Atlas数据库验证所筛选出的基因在转录组和蛋白表达水平上的差异。结果共筛选出5个关键差异表达基因,分别是AHSG、SERPINC1、CYP2E1、FGG及TTR,其主要的生物学过程富集在氧化还原反应、药物反应和跨膜转运等方面;信号通路富集在代谢途径、抗生素生物合成、补体途径和细胞色素P450系统等方面。结论利用生物信息学方法,通过不同数据平台的比较和验证,筛选出胆管癌差异表达的5个核心基因,并在转录和蛋白表达水平进行了验证,为胆管癌临床治疗提供了新的候选分子靶点。
Objective To identify the key genes and signaling pathways in the pathogenesis of cholangiocarcinoma and to provide data support for the search for therapeutic targets.Methods Two microarray datasets GSE26566 and GSE76297 of GEO database were used to screen the differential expression genes,and the functional enrichment analysis was carried out by DAVID database.The PPI network between gene coding proteins was constructed by STRING database,and then imported into Cytoscape software for visual analysis.The differences in transcriptional group and protein expression level of the screened genes were verified by TCGA and The Human Protein Atlas databases.Results Five key differentially expressed genes were screened:AHSG,SERPINC1,CYP2E1,FGG and TTR.Their main biological processes are enriched in redox reactions,drug reactions and transmembrane transport,while signal pathways are enriched in metabolic pathways,antibiotic biosynthesis,complement pathways and cytochrome P450 systems.Conclusion By using the method of bioinformatics,through the comparison and verification of different data platforms,differentially expressed genes were screened to provide new key molecular targets for the treatment of cholangiocarcinoma.
