摘要
采用开环聚合法制备PCL-PEG-PCL共聚物,并将其制成温敏性水凝胶,探究了PEG(聚乙二醇)相对分子质量及质量浓度对水凝胶温敏性的影响.水凝胶的相变温度由翻转小瓶法测定.通过FTIR、热分析仪和SEM等技术对其组成及结构进行表征.以疏水性姜黄素(Cur)为模型药物,制备出载Cur PCL-PEG-PCL水凝胶,并研究其体外释药行为.FTIR结果表明:实验制备的共聚物中含有PCL和PEG的链段.热分析结果表明:在25℃~65℃内水凝胶存在相变过程.SEM结果表明:水凝胶剖面具有疏松多孔.体外释药结果表明:PCL-PEG-PCL水凝胶对Cur具有缓释作用,释药机理符合Higuchi骨架溶蚀模型.
PCL-PEG-PCL polymers were prepared by ring-opening polymerization and made into thermosensitive hydrogel.Meanwhile,the effects of molecular weight and quality concentration of PEG on the thermosensitive hydrogel were also investigated.The sol-gel transition temperature was determined with the test tube inverting method.The composition and structure of the hydrogels were characterized by FTIR,TGA,SME analysis technology.A series of Cur-loaded PCL-PEG-PCL hydrogel were prepared with different concentrations of blank hydrogel as matrix and hydrophobic curcumin(Cur)as model drug.The drug release behavior of drug-loaded hydrogel was studied.FTIR results showed that the copolymers contained the segments of PCL and PEG.The results of thermal analysis showed that it’s a phase transition process at 25℃~65℃.SEM confirmed the structure of the hydrogel was loose and porous.In vitro release results shown that PCL-PEG-PCL hydrogel has sustained-release effect on Cur,and the drug release mechanism was consistent with the Higuchi skeleton dissolution models.
