摘要
目的研究乳腺癌癌组织中溶酶体四次跨膜蛋白质B(LAPTM4B)、dCTP焦磷酸酶(DCTPP1)的表达水平及临床意义。方法选取2014年3月至2016年6月本院收治的乳腺癌患者93例作为研究对象,收集患者手术切除的肿瘤组织以及癌旁正常组织,采用免疫组化法(SP)检测两组组织中LAPTM4B、DCTPP1的表达情况,分析其临床意义。结果乳腺癌癌组织部位LAPTM4B、DCTPP1阳性表达率明显高于癌旁组织,差异具有统计学意义(P<0.05);TNM分期为Ⅱ期的乳腺癌患者LAPTM4B、DCTPP1阳性率显著高于TNMⅠ期(LAPTM4B、DCTPP1阳性率和Ⅱ期患者(LAPTM4B、DCTPP1阳性率、淋巴结转移的乳腺癌患者LAPTM4B、DCTPP1阳性表达率低于无淋巴结转移患者(LAPTM4B、DCTPP1阳性率,差异均具有统计学意义(P<0.05);随访时间36个月,LAPTM4B、DCTPP1表达阳性患者3年生存率和生存时间均低于阴性患者的生存率和生存时间,差异具有统计学意义(P<0.05);LAPTM4B和DCTPP1同时表达阳性患者3年生存率显著低于两者均阴性患者以及仅LAPTM4B表达阴性或者仅DCTPP1表达阴性患者生存率和生存时间,差异均具有统计学意义(P<0.05)。结论乳腺癌患者LAPTM4B和DCTPP1水平呈高表达状态,且其表达阳性率与患者的TNM分期、淋巴结转移以及预后均存在密切联系,有望成为乳腺癌诊断和预后的评估指标。
Objective To study the expression levels of lysosome-associated protein transmembrane 4 beta(LAPTM4 B)and dCTP pyrophosphatase 1(DCTPP1)in breast cancer tissues and their clinical significance. Methods Ninety-three breast cancer patients who were admitted and treated in the hospital between March 2014 and June 2016 were selected as the study subjects. The tumor tissues and normal adjacent tissues surgically removed were collected to detect the expression of LAPTM4 B and DCTPP1 by immunohistochemical method(SP),and their clinical significance was analyzed. Results The positive expression rates of LAPTM4 B and DCTPP1 in breast cancer tissues were significantly higher than those in adjacent tissues(P<0.05). The positive expression rates of LAPTM4 B and DCTPP1 in patients with breast cancer at TNM stageⅡwere significantly higher than those at TNM stageⅠand stageⅡ. The positive expression rates of LAPTM4 B and DCTPP1 in breast cancer patients with lymph node metastasis were lower than those without lymph node metastasis(P<0.05). The follow-up time was 36 months. The 3-year survival rate and survival time of LAPTM4 B or DCTPP1 positive patients were lower/shorter than those of LAPTM4 B or DCTPP1 negative ones(P < 0.05). The 3-year survival rate and the survival time of both LAPTM4 B and DCTPP1 positive patients were significantly lower/shorter than those of neither LAPTM4 B nor DCTPP1 negative patients(P<0.05). Conclusion The levels of LAPTM4 B and DCTPP1 in breast cancer patients are highly expressed,and their positive expression rates are closely related to the TNM staging,lymph node metastasis and prognosis of the patients,and they are expected to become indicators of breast cancer diagnosis and prognosis.
